摘要
目的研究健脾消癌方对缺氧环境下SW620结肠癌细胞裸鼠移植瘤的抑制作用。方法皮下注射结肠癌SW620细胞建立裸鼠皮下移植瘤模型,采用含10.5%O2,89.5%N2的混合气体模拟缺氧微环境,将裸鼠随机分为健脾消癌方组和对照组,每组25只。健脾消癌方组予健脾消癌方灌胃,对照组予等体积的0.9%NaCl灌胃,连续给药28 d。观察裸鼠一般情况,体重和肿瘤生长情况,用苏木素—伊红染色法(HE)观察病理形态学的改变。结果健脾消癌方瘤体体积为(280.30±37.89)mm3,瘤体质量为(0.342±0.06)g,与对照组比较,瘤体体积及质量均明显降低(P<0.01,P<0.05);健脾消癌方组的抑瘤率为30.4%。结论在缺氧条件下,健脾消癌方能提高结肠癌模型裸鼠的生活质量,抑制SW620结肠癌细胞裸鼠移植瘤的生长。
Objective To study the effect of Jianpi Xiaoai Recipe(JXP)on the growth of transplanted SW620 colon cancer in nude mice under hypoxic microenvironment.Methods The mouse model was established by subcutaneous injection of colon cancer SW620 cells and by exposure to the hypoxic microenvironment with mixed gas containing10.5%O2 and 89.5%N2,and then the modeled mice were randomly divided into JXP group and control group,25 mice in each group.JXP group was treated with intragastric gavage of JXP,and the control group was treated with the same volume of physiological saline by intragastric gavage for 4 continuous weeks.The general status,body weight and tumor growth of nude mice were observed in the two groups.Hematoxylin-eosin staining(HE)was used to observe the pathological changes in the colon.Results The volume of the xenograft tumor was(280.30±37.89)mm3 and the average weight of the xenograft tumor was(0.342±0.06)g in JXP group,which were much less than those in the control group(P<0.01,P<0.05).The tumor-inhibition rate in JXP group was 30.4%.Conclusion Under hypoxic microenvironment,JXP can improve the quality of life of nude mice with colon cancer,and inhibit the growth of the xenograft tumor in nude mice with transplanted SW620 colon cancer.
