摘要
目的:研究江南卷柏药材与混淆品旱生卷柏、兖州卷柏的性状与HPLC特征图谱的区别,为江南卷柏和近缘种的鉴定寻找解决方案。方法:观察比较江南卷柏与混淆品的性状特征。采用HPLC法,利用相似度软件计算其HPLC谱图的相似度,选择具有鉴别意义的指纹区,以穗花杉双黄酮色谱峰为参照,计算色谱峰相对保留时间,建立特征图谱,采用HPLC-DAD-ESI-MS/MS指认江南卷柏特征图谱的化学成分,并与其近缘种图谱进行比较。结果:江南卷柏、旱生卷柏、兖州卷柏具有其专属的性状特征。HPLC特征图谱方法学考察良好,10批江南卷柏样品的相似度为0.871~0.985,具有稳定的共有峰。江南卷柏特征图谱中13种黄酮类化学成分得到指认,江南卷柏特征图谱与其分类关系最近的兖州卷柏、旱生卷柏的特征图谱有明显区别。罗布斯塔双黄酮、罗布斯塔双黄酮-4′-甲醚为首次从旱生卷柏中发现。结论:该方法简便,可靠,实用性强,能准确地鉴别江南卷柏以及近缘混淆品,为江南卷柏的鉴定提供了科学依据。
Objective:To study the difference of morphological characteristics and HPLC typical chromatogram of Selaginella moellendorffii and adulterants Selaginella stauntoniana,Selaginella involvens,so as to find a solution for identification of Selaginella moellendorffii and its related species.Methods:Morphological characteristics of Selaginella moellendorffii and adulterants were abserved and compared.HPLC method was used,similarity software was used to calculate the similarity of its HPLC chromatogram,fingerprint regions with identification significance were selected,amentoflavone chromatographic peak was taken as reference,relative retention time of chromatographic peak was calculated,and the characteristic chromatogram was established.Chemical components of the characteristic chromatogram of Selaginella moellendorffii were identified by HPLC-DAD-ESI-MS/MS.And the chromatogram of its related species was compared.Results:Selaginella moellendorffii,Selaginella stauntoniana and Selaginella involvens had their own characteristics.HPLC chromatogram method was well investigated.The similarity of 10 batches of Selaginella moellendorffii samples ranged from 0.871 to 0.985,with stable common peaks.The chemical constituents of 13 kinds of flavonoids of Selaginella moellendorffii were identified.The characteristic chromatogram of Selaginella moellendorffii was obviously different from that of Selaginella stauntoniana and Selaginella involvens,which had the closest taxonomic relationship.Robusta diflavone and 4′-methoxy robusta-flavone were first discovered from Selaginella stauntoniana.Conclusion:This method is simple,reliable and practical,and can accurately identify Selaginella moellendorffii and its related adulterants,which provide a scientific basis for identification of Selaginella moellendorffii.
