摘要
目的:为四川人工僵蚕生产和质量控制提供参考。方法:用时间-剂量-死亡率模型进行球孢白僵菌(BJ-MJ)对家蚕的毒力分析;分别测定不同死亡时间人工僵蚕样品、不同条重分组的自然僵蚕样品、桑叶、蚕砂的质量指标。结果:毒力测定表明BJ-MJ菌接种家蚕适宜的孢子悬浮液浓度为10^(5)~10^(6)个/mL。人工僵蚕与自然僵蚕的芦丁和白僵菌素含量接近。人工僵蚕浸出物含量符合中国药典规定,自然僵蚕除“吐丝缩短蚕体”浸出物含量低于20.0%,其余分组符合中国药典规定。人工僵蚕死亡时间早的(接种后4-6 d)样品总灰分高于7.0%,上蔟至吐丝阶段死亡的人工僵蚕总灰分符合中国药典规定;自然僵蚕中条重“0.40~0.49 g”(死亡时间早)的样品总灰分高于8.0%,蚕体完整硬实的自然僵蚕总灰分符合中国药典规定。桑叶总灰分含量高于9.0%,浸出物含量高于20%;蚕砂总灰分含量高于15.0%,浸出物含量10%左右;僵蚕体内的桑叶(或蚕砂)占比多总灰分含量高、僵蚕体内的桑叶占比多浸出物含量高。结论:人工僵蚕接种后死亡时间在第7天至吐丝阶段样品,丝腺发达,浸出物、总灰分符合中国药典标准,白僵菌素含量高,是人工生产僵蚕适宜的死亡时间。
Objective:To provide reference for the production and quality control of artificial Bombyx Batryticatus in Sichuan.Methods:The virulence analysis of Beauveria bassiana(BJ-MJ)to Bombyx mori was carried out by time-dose-mortality model.The quality indexes of artificial Bombyx Batryticatus samples with different death times and natural Bombyx Batryticatus samples with different weight groups,mulberry leaves and silkworm excrement were determined.Results:The virulence test results showed that the suitable concentration of spore suspension of Bombyx mori inoculated BJ-MJ was 10~5 to 10~6/mL.The contents of rutin and beauvericin in artificial Bombyx Batryticatus and natural Bombyx Batryticatus were similar.The extract content of artificial Bombyx Batryticatus conformed to the Chinese pharmacopoeia standard.The extract content of natural Bombyx Batryticatus except the"spinning shortened silkworm body"was less than 20.0%,the other groups were in accordance with the Chinese pharmacopoeia.The total ash content of natural Bombyx Batryticatus with early death time(4 to 6 days after inoculation)was higher than 7.0%,the total ash content of the dead Bombyx Batryticatus in the period from cluster to silking met the requirements of the Chinese pharmacopoeia;The total ash content of natural Bombyx Batryticatus with the weight of 0.40 to 0.49 g(early death time)was higher than 8.0%.The total ash content of natural Bombyx Batryticatus with complete and hard body met the requirements of Chinese pharmacopoeia.The total ash contents of mulberry leaves and silkworm excrement were higher than 9.0%,15.0%,respectively,the extract content of mulberry leaves was higher than 20%,while extract concent of silkworm excrement was about 10%;When the proportion of mulberry leaves(or silkworm excrement)in the body of Bombyx Batryticatus was high,the total ash content was high,while the proportion of mulberry leaves in the body of Bombyx Batryticatus was high,the extract content was high.Conclusion:The artificial Bombyx Batryticatus from the 7 th day of death time to silking stage after inoculating has well-developed silk glands,the extract and total ash contents are in line with the standard of Chinese pharmacopoeia,the content of beauvericin is high,it is the appropriate time of death for artificial production of Bombyx Batryticatus.
