摘要
目的:通过测定不同炮制品组方五子衍宗丸中浸出物及绿原酸、鞣花酸、金丝桃苷、异槲皮苷、毛蕊花糖苷、紫云英苷、山柰酚、五味子醇甲的含量,研究盐制对五子衍宗丸中浸出物及8个成分含量的影响。方法:按照2020年版中国药典四部2201项下方法测定水溶性及醇溶性浸出物含量;采用HPLC法测定样品中上述8个成分的含量。结果:五子衍宗丸中药物炮制后组方水溶性及醇溶性浸出物的含量明显增加,增加程度为盐制组>药典组。炮制后组方没有新的化学成分出现,但峰面积发生变化;药典组及盐制组绿原酸、金丝桃苷、异槲皮苷、毛蕊花糖苷的含量升高,紫云英苷、山柰酚、五味子醇甲的含量降低;盐制组鞣花酸的含量升高,但药典组鞣花酸的含量降低。结论:五子衍宗丸药物炮制后组方能促进饮片中化学成分的溶出,增加复方浸出物的含量;所建立的含量测定方法稳定、可靠,8个成分含量的变化可为研究五子衍宗丸中药物炮制前后组方化学成分、药理药效提供一定的依据。
Objective:To study the effects of salt-water processing on the contents of Wuzi yanzong pills extracts and 8 components by determining the contents of extracts,chlorogenic acid,ellagic acid,hyperoside,isoquercitrin,verbascoside,astragalin,kaempferol,schisandrin in Wuzi yanzong pills with different processing products.Methods:The water soluble and alcohol soluble extracts contents were determined according to the method under item 2201 of the fourth part of Chinese Pharmacopoeia(2020 edition).The contents of the above 8 components in the samples were determined by HPLC.Results:After drug processing in Wuzi yanzong pills,the contents of water soluble and alcohol soluble extracts increased obviously,and the increase degree was salt-water processing group>pharmacopoeia group.After processing,no new chemical composition appeared,but the peak area changed.The contents of chlorogenic acid,hyperoside,isoquercitrin and verbascoside in pharmacopoeia group and salt-water processing group were increased,while the contents of astragalin,kaempferol and schisandrin were decreased.The content of ellagic acid was increased in salt-water processing group,but decreased in pharmacopoeia group.Conclusion:The drug processing of Wuzi yanzong pills can promote the dissolution of chemical components and increase the content of compound extract.The established method is stable and reliable,and the changes of the contents of 8 components can provide some basis for the study of the chemical composition and pharmacological effect of Wuzi yanzong pills before and after the drug processing.
