基于UHPLC-Q-Orbitrap HRMS的羊红膻血清药物化学分析 被引量:3

Study on Serum Pharmacochemistry of Pimpinella thellungiana Whole Herb with Roots by UHPLC-Q-Orbitrap HRMS

摘要 目的:对羊红膻药材进行血清药物化学研究,分析其体内入血成分。方法:采用超高效液相色谱-四级杆-轨道肼高分辨质谱联用(UHPLC-Q-Orbitrap HRMS)技术,通过比对羊红膻药材、空白血清和含药血清的图谱差异,解析羊红膻水提液经大鼠灌胃后血清中的原型成分和代谢产物。结果:给药后,从血清中鉴定了43个吸收入血成分,其中19个为原型成分,24个为原型成分的代谢产物。入血的原型成分主要为咖啡酰奎宁酸类、黄酮类和小分子酚酸类。结论:该研究较为全面地阐释了羊红膻水提液在大鼠血中的移行成分,为进一步研究其药效物质和作用机制奠定基础。 Objective:To study the serum pharmacochemistry of Pimpinella thellungiana whole herb with roots,and to analysis its components in the blood.Methods:Ultra high performance liquid chromatography coupled with Q-exactive focus hybrid quadrupole and orbitrap high resolution mass spectrometer(UHPLC-Q-Orbitrap HRMS)technique was used to analyze the prototypical components and metabolites of water extract from Pimpinella thellungiana whole herb with roots after gavage in rats by comparing the chromatogram differences of Pimpinella thellungiana whole herb with roots,blank serum and drug-containing serum.Results:After administration,43 absorbed components were identified from serum,among which 19 were prototype components and 24 were metabolites of prototype components.The prototypical components in the blood were mainly caffeoquinic acid,flavonoid and small molecule phenolic acid.Conclusion:This study comprehensively elucidates the migration components of water extract of Pimpinella thellungiana whole herb with roots in rat blood,which lays the foundation for further study of its pharmacodynamic substance and mechanism.
