摘要
目的:通过药效学实验筛选出活血消瘿片的提取工艺路线,并通过正交试验法优选出最佳的活血消瘿片提取工艺参数。方法:以不同提取方式的活血消瘿片开展药效学实验,以SD大鼠的甲状腺的质量、游离三碘甲状腺原氨酸(FT3)、游离甲状腺素(FT4)、第三代促甲状腺激素(TSH3)为药效指标,筛选出最佳提取工艺方式,以处方中多个化学成分的测定和干膏率为评价指标,通过正交试验筛选出最佳工艺参数,并进行验证。结果:药效学实验结果表明,处方直接水提为最佳提取方式,以加入12倍量水,加热提取3次,每次提取2 h为最佳的提取工艺参数。结论:活血消瘿片能有效减轻因丙硫氧嘧啶引起的甲状腺肿大,影响FT3、FT4、TSH3的变化。优选的最佳提取工艺参数合理,为后续的产业化开发奠定研究基础。
Objective:To select the extraction method of Huoxue xiaoying tablets by pharmacodynamics experiment,to select the best extraction process parameters of Huoxue xiaoying tablets by orthogonal test.Methods:Pharmacodynamics experiment was carried out on Huoxue xiaoying tablets with different extraction methods.The best extraction process was selected with the quality of thyroid,free triiodothyronine(FT3),free thyroxine(FT4)and third generation thyroid stimulating hormone(TSH3)in SD rats as pharmacodynamic indexes.The determination of several chemical components in the prescription and the rate of dry ointment were used as evaluation indexes,the optimum technological parameters were selected by orthogonal experiment and verified.Results:The results of pharmacodynamics experiment showed that water extraction was the best extraction method,the optimum extraction parameters were as follows:adding 12 times of water,heating extraction for 3 times,2 hours each time.Conclusion:Huoxue xiaoying tablets can effectively reduce goiter caused by propylthiouracil and affect the changes of FT3,FT4,TSH3.The optimized extraction process parameters are reasonable,which lays a research foundation for subsequent industrial development.
