藤茶双氢杨梅素对肝纤维化小鼠TGF-β1/Smad信号通路的影响 被引量:12

Effects of Dihydromyricetin from Ampelopsis grossedntata on TGF-βSmad Signaling Pathway in Hepatic Fibrosis Mice

摘要 目的:探究藤茶双氢杨梅素抗肝纤维化的作用机制。方法:昆明种小鼠60只(雌雄各半)随机分为正常对照组、模型组、秋水仙碱阳性对照组及双氢杨梅素高、中、低剂量组。除正常对照组外,各组小鼠背部皮下注射25%CCl4花生油溶液,1次/3 d,连续10 w,造模同时各组小鼠灌胃相应药物,正常对照组灌胃蒸馏水,1次/d,连续10 w。实验结束后检测血清丙氨酸氨基转移酶(ALT)、门冬氨酸氨基转移酶(AST)及肝组织胶原蛋白Ⅰ(COLⅠ)、胶原蛋白Ⅲ(COLⅢ)水平,并检测肝纤维化程度、肝组织TGF-β1/Smad信号通路主要蛋白及mRNA的表达。结果:与模型组比较,双氢杨梅素可降低肝纤维化小鼠血清ALT、AST及肝组织COLⅠ、COLⅢ水平,减少肝脏胶原蛋白增生,下调肝组织TGF-β1、Smad2、Smad4、α-SMA mRNA及蛋白表达,上调Smad7 mRNA及蛋白表达。结论:双氢杨梅素抗肝纤维化的机制可能与调节TGF-β1/Smad信号通路有关。 Objective:To explore the mechanism of dihydromyricetin from Ampelopsis grossedntata against liver fibrosis.Methods:Sixty Kunming mice(half male and half female)were randomly divided into six groups,including normal control group,model group,colchicine positive control group,dihydromyricetin high-,middle-,low-dose groups.Expect normal control group,mice in each group were subcutaneously injected with 25% CCl4 peanut oil solution on the back of each group for 1 time/3 d,10 w continuously.Meanwhile,mice in each group were given corresponding drugs.Mice in the normal control group were given distilled water for 1 time/d,10 w continuously.At the end of the experiment,the serum alanine aminotransferase(ALT),aspartate aminotransferase(AST)and liver tissue collagen Ⅰ(COL Ⅰ),collagen Ⅲ(COL Ⅲ)levels,the degree of liver fibrosis,main protein and gene expressions of liver tissue TGF-β1/Smad signaling pathways were detected.Results:Compared with the model group,dihydromyricetin could decrease the levels of ALT and AST in serum and COL Ⅰ and COL Ⅲ in liver tissue,decrease the proliferation of collagen in liver,down-regulate the expressions of TGF-β1,Smad2,Smad4,α-SMA protein and gene in liver tissue,up-regulate the expressions of Smad7 mRNA and protein.Conclusion:The mechanism of dihydromyricetin against liver fibrosis may be related to the regulation of TGF-β1/Smad signaling pathway.
