摘要
目的:研究环维黄杨星D对醛固酮(ALD)联合高盐诱导大鼠实验性心衰模型心功能障碍的改善作用。方法:采用ALD皮下注射联合高钠盐饮水建立大鼠实验性心衰模型,实验分为正常对照组、模型组及环维黄杨星D低(1.1 mg/kg)、高(2.2 mg/kg)剂量组,造模4 w后监测血流动力学指标并分析心功能的变化;Elisa法检测血浆ALD和可溶型ST2(sST2)水平;Masson染色观察心肌重塑程度;计算大鼠心室重塑参数。结果:ALD皮下注射联合高钠盐饮水4 w后,与正常对照组比较,模型大鼠HR、SBP、DBP、MAP、LVSP、+dp/dtmax、-dp/dtmax均显著降低,LVEDP、HL/LVD、LVAW/LVD及血浆中ALD、sST2含量显著升高;与模型组比较,环维黄杨星D各组上述指标均显著改善。Masson染色可见ALD皮下注射联合高钠盐饮水4 w后,大鼠心脏中大量胶原纤维沉积、残留心肌数目少、心肌组织排列紊乱;与模型组比较,环维黄杨星D各组可见心肌重塑部分改善。结论:环维黄杨星D对ALD联合高盐诱导大鼠实验性心衰模型心功能具有改善作用。
Objective:To investigate the amelioration effect of cyclovirobuxine D(CVB-D)on cardiac dysfunction in chronic heart failure rat induced by aldosterone(ALD)combined with high salt.Methods:The rat model of heart failure was established by hypodermic injection of ALD with high sodium salt water.The experiment was divided into normal control group,model group,CVB-D low dose group(1.1 mg/kg)and high dose group(2.2 mg/kg).The hemodynamic parameters were measured after 4 w and the alterations of cardiac function were analyzed.Elisa method was used to detect plasma ALD and sST2 levels;Masson staining was used to observe the degree of myocardial remodeling changes;Rat ventricular remodeling parameters were calculated.Results:Compared with normal control group,the HR,SBP,DBP,MAP,LVSP,+dp/dtmax and-dp/dtmaxof model rats were significantly decreased after hypodermic injection of ALD combined with high sodium salt diet for 4 w,LVEDP,HL/LVD,LVAW/LVD and plasma ALT,sST2 contents significantly increased.Compared with model group,the above indexes in each CVB-D group were significantly improved.Masson staining showed that a large number of collagen fibers were deposited in the model group with fewer cardiac muscleresidues and myocardial tissue was disorganized.Compared with model group,these myocardial remodeling were partially ameliorated by CVB-D.Conclusion:Cyclovirobuxine D can improve the cardiac dysfunction in rats with experimental heart failure induced by ALD combined with high salt.
