Qualitative Analysis on Chemical Constituents from Epimedium brevicornu by UPLC-Q-TOF-MS/MS

摘要 目的:采用超高效液相色谱-飞行时间质谱联用技术(UPLC-Q-TOF-MS/MS)对淫羊藿中的化学成分进行定性分析。方法:采用CORTECS C18色谱柱(100 mm×2.1 mm,1.6μm),以0.1%甲酸溶液(A)-乙腈(B)为流动相进行梯度洗脱,流速为0.3 mL/min,柱温30℃,进样量5μL。质谱采用电喷雾(ESI)离子源,正、负离子模式监测,通过保留时间、相对分子质量、二级质谱碎片等信息,结合对照品碎片裂解规律及文献数据对检测到的化学成分进行定性分析。结果:共鉴定和推测出淫羊藿中47个化学成分,包括39个黄酮类、5个有机酸类、3个其他类成分。结论:该UPLC-Q-TOF-MS/MS方法能快速、准确地分析淫羊藿中的化学成分,为提高其质量控制水平和阐明药效物质基础提供了参考。 Objective:To analyze and identify the chemical constituents from Epimedium brevicornu by UPLC-Q-TOF-MS/MS.Methods:Using the CORTECS C18 chromatographic column(100 mm×2.1 mm,1.6 μm)with the temperature of 30 ℃,the mobile phase was 0.1% formic acid solution(A)and acetonitrile(B)with gradient elution at a flow rate of 0.3 mL/min,the injection volume was 5 μL.The MS spectrum was acquired in both negative and positive ion modes using ESI ion source.The compounds were identified by retention time,relative molecular mass and secondary mass spectrometry pyrolysis fragments,as well as fragmentation regularity summarized from reference substance and the literature.Results:Fourty-seven compounds from Epimedium brevicornu were successfully identified and predicted,including thirty-nine flavonoids,five organic acids and three other types.Conclusion:UPLC-Q-TOF-MS/MS method can rapidly and accurately analyze the chemical components in Epimedium brevicornu,it provides reference for improving the quality control level and clarifying the basis of medicinal materials.
