酶辅助提取畲药三叶青总三萜的工艺研究 被引量:2

摘要 目的:确定酶辅助优化畲药三叶青总三萜提取工艺参数。方法:采用紫外分光光度法对三叶青总三萜进行测定,以总三萜提取率为评价指标。在单因素试验基础上,以酶的种类、酶解pH值、酶解温度、酶解时间、酶添加量为考察因素,通过L9(34)正交试验优化三叶青总三萜提取工艺。结果:酶解提取三叶青总三萜的最佳工艺参数为:纤维素酶添加量为2.5%、酶解温度为45℃、酶解时间为90 min、酶解pH值为5。在此工艺条件下,经放大验证试验,三叶青总三萜提取率为(2.74±0.07)%,RSD为2.17%。结论:正交实验优化的酶辅助提取三叶青总三萜工艺参数合理,可为三叶青三萜类成分的提取利用提供参考。 Objective:To determine the enzyme-assisted optimization of the extraction process parameters of total triterpenoids from She medicine Trifolium.Methods:The total triterpenoids of Ilex tricolor were determined by ultraviolet spectrophotometry,and the extraction rate of total triterpenoids was used as the evaluation index.On the basis of single factor test,the extraction process of total triterpenoids from Ilex trifoliata was optimized by L9(34)orthogonal test with the type of enzyme,pH value of enzyme hydrolysis,temperature of enzyme hydrolysis,time of enzyme hydrolysis,and amount of enzyme added as the investigation factors.Results:The optimum technological parameters for enzymatic extraction of total triterpenes from Ilex tricolor are as follows:the addition of cellulase is 2.5%,the temperature of enzymatic hydrolysis is 45℃,the time of enzymatic hydrolysis is 90 min,and the pH value of enzymatic hydrolysis is 5.Under this technological condition,the extraction rate of total triterpenoids from Ilex tricolor was(2.74±0.07)%,and the RSD was 2.17%.Conclusion:The optimum parameters of enzyme-assisted extraction of total triterpenoids from Ilex tricolor by orthogonal experiment are reasonable,which can provide reference for the extraction and utilization of triterpenoids from Ilex tricolor.
