摘要
目的探讨AKR1B10蛋白在结肠癌组织中的表达,并分析与其临床病理特征的相关性。方法选取2017年1月至2018年12月间辽宁省锦州医科大学第一附属医院收治的70例结肠癌患者的临床资料及结肠癌组织标本,分别采用反转录聚合酶链反应(RT-PCR)、蛋白质印迹法和免疫组织化学法检测结肠癌组织及癌旁组织中AKR1B10 mRNA及蛋白的表达情况,并分析其与结肠癌病理特征的关系。结果结肠癌组织中AKR1B10 mRNA相对表达量为(1.73±0.06),高于其癌旁正常组织(0.99±0.03),差异有统计学意义(P<0.05)。结肠癌AKR1B10蛋白光密度壁纸亦显著高于癌旁组织,差异有统计学意义(P<0.01)。结肠癌组织和正常组织基底层细胞区均有AKR1B10的表达,其表达主要在细胞质,亦可见细胞核的表达。其中在70例结肠癌组织中,61例(87.1%)AKR1B10蛋白表达阳性,而在70例癌旁组织中,22例(31.4%)AKR1B10的阳性表达,差异有统计学意义(P<0.05)。结肠癌组织中AKR1B10的表达与患者肿瘤大小、肿瘤浸润深度及淋巴结转移均有关,差异均有统计学意义(均P<0.05)。结论AKR1B10在结肠癌组织中呈过度表达,且表达情况与TMN分期和淋巴结转移等临床病理特征存在相关性,可能成为结肠癌的治疗靶点之一。
Objective To detect the expression of AKR1 B10 in colon cancer tissues and to analyze its relationship with clinicopathological features.Methods The clinical data and tissue samples were selected in 70 patients with colon cancer who admitted to The First Affiliated Hospital of Jinzhou Medical University from January 2017 to December 2018.The expression of AKR1 B10 mRNA and protein in colon cancer tissues and paracancerous tissues were detected by reverse transcriptase polymerase chain reaction(RT-PCR),Western blot and immunohistochemistry,respectively.The relationship between the expression of AKR1 B10 and the pathological features of colon cancer was analyzed.Results The expression level of AKR1 B10 mRNA was 1.73±0.06 in colon cancer tissues which was higher than 0.99±0.03 in normal paracancerous tissues(P<0.05).Optical density for AKR1 B10 protein was higher in colon cancer tissues than in normal paracancerous tissues(P<0.05).The expression of AKR1 B10 was also observed in substrate area both in the colon cancer tissues and in the normal paracancerous tissues(all P<0.05).The expression of AKR1 B10 was primarily observed in cytoplasm followed by nucleus.Of 70 colon cancer tissues,61(87.1%)tissues were observed to have positive expression of AKR1 B10 protein.Comparatively,positive expression of AKR1 B10 protein was observed in 22(31.4%)tissues among 70 normal paracancerous tissues(P<0.05).AKR1 B10 expression was associated with tumor size,tumor infiltration depth,and lymph node metastasis(all P<0.05).Conclusion AKR1 B10 is overexpressed in colon cancer tissues,and its expression is correlated with TNM staging,lymph node metastasis and other clinicopathological features,suggesting that it may become one of the therapeutic targets for colon cancer.
