摘要
目的分析浙江省临海市2016-2019年禽流感病毒(avian influenza virus,AIV)外环境主要亚型和分布特点。方法在临海市40个涉禽场所随机采集5类外环境样本514份,采用实时RT-PCR方法检测通用甲型流感病毒(influenza A virus,FluA),阳性样本进一步进行H5、H7、H9亚型检测,采用SPSS22.0软件进行统计分析。结果样本中Flu A阳性率达47.86%,H5亚型、H7亚型、H9亚型及未分型总阳性率分别为2.92%、7.78%、31.13%、10.70%,差异有统计学意义(χ~2=249.599,P=0.000),存在H5+H9(1.95%)和H7+H9(2.72%)混合型;每年一、四季度Flu A阳性率最高,达37.78%~60.00%和20.00%~73.68%;不同年份Flu A阳性率差异有统计学意义(χ~2=11.226,P=0.011);不同涉禽场所中,农贸市场的Flu A核酸阳性率(53.72%)高于养殖场(12.31%)(χ~2=38.931,P=0.000);不同类型样本中,禽类饮用水样本Flu A核酸阳性率最高(76.29%),其次是清洗禽类的污水(68.00%),再是粪便样本(43.22%)。结论临海市涉禽场所外环境普遍存在禽流感病毒污染,H9亚型是主要的病原体,混合型占一定比例,存在其他亚型,污染高峰为每年的第一、第四季度。
Objective To understand the major subtypes and distribution of avian influenza A virus in poultry-related environment in Linhai City of Zhejiang province during 2016 and 2019.Methods A total of 514 environmental samples of 5 major categories were randomly collected from 40 poultry related sites for the detection of influenza A virus(FluA)by real-time PCR and subtype H5,H7 and H9 were further determined by sequencing.Data were statistically analyzed using SPSS 22.0.Results The overall positive rate of Flu A virus was 47.86%,and the positive rates of subtype H5,H7,H9 and undetermined subtype were 2.92%,7.78%,31.13%and 10.70%,respectively,the difference was statistically significant(χ~2=249.599,P=0.000).1.95%and 2.72%of samples were found double positive for H5+H9+and H7+H9+.The positive rates of influenza A virus in the first quarter and fourth quarter were the highest,ranging from 37.78%to 60.00%and from 20.00%to 73.68%,respectively.The difference in annual positive rate of avian influenza A virus was statistically significant(χ~2=11.226,P=0.011).The positive rate was higher in farmers markets than that in poultry farms(53.72%vs 12.31%,χ~2=38.931,P=0.000).The RNA positive rate was the highest of 76.29%in poultry drinking water,followed by 68.00%in poultry sewage and 43.22%in fecal samples.Conclusions Avian influenza virus contamination is popular in the poultry related environment in Linhai City,especially in the first and fourth quarters of the year with the predominant subtype of H9.Co-contamination with H5 and H9 or H7 and H9 and other subtypes also exist.Management of the poultry farms should be strengthened;disinfection and cleaning of poultry drinking water and poultry sewage should be implemented effectively.
