摘要
锰离子作为一种微量元素在机体内分泌、免疫系统、造血功能等方面起着重要的调节作用。近年来锰离子被发现作为cGAS-STING通路的调节剂,一方面强化cGAS对DNA的敏感度促进cGAMP的合成,另一方面直接结合并激活STING蛋白。鉴于cGAS-STING通路在肿瘤免疫治疗方面的作用,锰离子在肿瘤免疫治疗方面的潜力引起了人们的注意。文章综述了锰离子在肿瘤治疗方面的应用及作用原理,着重总结了近些年来基于Mn^(2+)对cGAS-STING通路激活作用的锰免疫疗法,包括多种锰及含锰制剂的单独使用及联合治疗策略。
Manganese ion as a trace element plays an important regulatory role in the endocrine,immune system and hematopoietic functions of the body.In recent years,manganese ion has been found to act as a regulator of the cGAS-STING pathway,which on the one hand enhances the sensitivity of cGAS to DNA to promote the synthesis of cGAMP,and on the other hand directly binds and acti-vates STING proteins.cGAS-STING pathway is one of the popular targets for tumor immunotherapy in recent years,and these findings suggest the potential of Mn^(2+)in tumor immunotherapy.This article reviews the progress of intrinsic immunity and important intrinsic immune pathway cGAS-STING-related therapies in tumor immunotherapy,followed by a summary of manganese immunotherapy based on the activation of cGAS-STING pathway by Mn^(2+)in recent years,including the use of various Mn and Mn-containing agents alone and in combination therapeutic strategies.
