摘要
目的:《中国药典》2020版第一部"续断鉴别及含量测定"项下的方法较为复杂,应重新建立续断的薄层鉴别方法及川续断皂苷Ⅵ的含量测定方法。方法:采用薄层鉴别法(TLC)对续断及其有效成分川续断皂苷Ⅵ进行鉴别。建立高效液相色谱法(HPLC)测定川续断皂苷Ⅵ,色谱柱为C_(18)(250mm×4.6mm,5μm),柱温35℃,流动相为乙腈∶水(30∶70),体积流量1mL/min;检测波长212nm。结果:薄层鉴别方法具有较好的分离度及较强的专属性。川续断皂苷Ⅵ在一定范围内与峰面积呈良好的线性关系,平均回收率为101.74%(RSD=1.16%)。结论:该方法简便、准确、重现性好,可用于续断的薄层鉴别及川续断皂苷Ⅵ的含量测定。
Objective:The method under“identification and content determination of Dipsaci Radix”in the first part of Chinese Pharmacopoeia 2020 edition is complex.The TLC identification method of Dipsaci Radixand the content determination method of AsperosaponinⅥshould be re established.Methods:Dipsaci Radixand its effective component AsperosaponinⅥwere identified by TLC.An HPLC method was established for the determination of AsperosaponinⅥwith C_(18)(250 mm column×4.6 mm,5μm),column temperature 35℃,mobile phase acetonitrile:water(30∶70),volume flow rate 1 mL/min;The detection wavelength is 212 nm.Results:The TLC method had good resolution and strong specificity.The average recovery of AsperosaponinⅥwas 101.74%(RSD=1.16%).Conclusion:The method is simple,accurate and reproducible.It can be used for TLC identification of Dipsaci Radixand determination of Asperosaponin Ⅵ.
