摘要
目的:建立百合药材王百合苷B含量测定方法及UPLC特征图谱测定方法,考察不同初加工方法对百合药材质量的影响。方法:采用直接烘干法、冷水浸泡法、烫制法及蒸制法对百合鲜品进行初加工,以性状、水分、浸出物、王百合苷B含量及特征图谱为质量评价指标,优选初加工方法。结果:不同初加工方法对百合药材的质量影响较大,烫制、蒸制百合样品的王百合苷B含量显著高于直接烘干及冷水浸泡百合样品的王百合苷B含量;18批百合药材的特征图谱均呈现7个特征峰,直接烘干及冷水浸泡百合样品的峰4均丢失,烫制及蒸制百合样品无特征峰丢失;直接烘干、冷水浸泡、烫12、16min及蒸16min百合样品的外观不符合要求,其他样品外观符合要求;4种初加工方法百合样品的水分与浸出物无明显差异。结论:建立的王百合苷B含量测定方法与UPLC特征图谱测定方法可用于百合药材的质量评价,百合适宜的初加工方法为沸水烫制4~8min或蒸制4~12min。
Objective:To establish a method for the determination of regaloside B and UPLC characteristic chromatogram of Lilii Bulbus,and to investigate the effects of different initial processing methods on the quality of Lilii Bulbus.Methods:Direct drying,cold water soaking,scalding and steaming methods were used for the initial processing of Lilii Bulbus fresh products.The appearance,moisture,extraction,content of regaloside B and characteristic chromatogram were used as quality evaluation indexes to optimize the initial processing methods.Results:Different initial processing methods had a greater effect on the quality of Lilii Bulbus.The content of regaloside B in scalding and steaming Lilii Bulbus samples was significantly higher than the content of regaloside B in the direct drying and cold water soaking Lilii Bulbus samples;The characteristic chromatogram of 18 batches of Lilii Bulbus were presented7 characteristic peaks,and the peaks 4 of the direct drying and cold water soaking Lilii Bulbus samples were lost,while no characteristic peaks of the scalding and steaming Lilii Bulbus samples were lost;The appearance of Lilii Bulbus samples direct drying,soaking in cold water,scalding for 12、16 min and steaming for 16 min did not meet the requirements,while the appearance of other samples met the requirements.There were no significant differences in the moisture and extraction of the Lilii Bulbus samples in the four initial processing methods.Conclusion:The method of determination of regaloside B and UPLC characteristic chromatogram established in this study can be used for the quality evaluation of Lilii Bulbus.The suitable initial processing method of Lilii Bulbus is scalding for 4 to 8 min or steaming for 4 to 12 min.
