摘要
目的:应用网络药理学的方法预测苗药黑骨藤治疗类风湿性关节炎(RA)的作用机制。方法:利用中药系统药理学(TCMSP)数据库、CNKI、PubMed以及文献查阅的方法筛选黑骨藤活性成分,在Swiss Target Prediction数据库中预测活性成分的靶点,采用Genecards数据库、OMIM数据库检索RA疾病靶点,构建"药物-成分-疾病-靶点"网络图,构建核心靶点的蛋白-蛋白互相作用(PPI)网络图,并进行基因本体(GO)生物过程与KEGG通路富集分析。结果:检索出黑骨藤治疗RA的活性成分10个,主要为甾类及萜类,药物-疾病的共同靶点352个,如AKT1、IL-6、VEGF、MAPK3等,涉及细胞凋亡、一氧化氮(NO)合成、血小板激活、蛋白磷酸化、细胞增殖、T细胞聚集等多个GO生物过程,以及PI3K-AKT、ErbB、催乳素、雌激素、VEGF等多个信号通路。结论:黑骨藤可能通过介导PI3K-AKT、ErbB、雌激素、VEGF等信号通路参与抑制细胞凋亡、促进细胞增殖、促进NO合成、抑制炎症反应等过程,发挥抗RA作用。
Objective:Predict the mechanism of Miao medicine Periploca forrestii Schltr in the treatment of Rheumatoid Arthritis(RA)with the method of network pharmacology.Methods:The active componentsof Miao medicine Periploca forrestii Schltr were screened by using the TCMSP database,CNKI,PubMed and literature review,and the target of the active components was predicted in the Swiss Target Prediction database,using Genecards database and Omim database to search for RA disease targets,construct"drug-ingredient-disease-target"network graph,construct Protein-Protein Interaction(PPI)network graph of core target,gene ontology(GO)bioprocess and KEGG pathway enrichment analysis were performed.Results:There were 10 active components,mainly Steroids and Terpenoids,and 352 common targets of drug-disease,such as AKT1,IL-6,VEGF,MAPK3,etc.Including Cell Apoptosis,Nitric Oxide synthesis,platelet activation,protein phosphorylation,cell proliferation,T cell aggregation and other go biological processes and PI3 K-AKT,ErbB,prolactin,estrogen,VEGF signaling pathways.Conclusion:Miao medicine Periploca forrestii Schltr may play an anti-RA role by mediating PI3 K-AKT,ErbB,estrogen,VEGF and other signaling pathways in the process of inhibiting cell apoptosis,promoting cell proliferation,promoting NO synthesis and inhibiting inflammatory reaction.
