风湿性心脏瓣膜病合并心房颤动患者右心耳中超级化激活环核苷酸调控通道2和4的协同表达

Coordinated expression of hyperpolarization-activated cyclic nucleotide-gated channel-2 and hyperpolarization-activated cyclic nucleotide-gated channel-4 in atrial fibrillation patients with rheumatic heart disease

摘要 目的探讨超级化激活环核苷酸调控通道-2(HCN2)和HCN4在风湿性心脏瓣膜病(RHD)患者右心耳组织的协同表达。方法收集2019年1月至2020年12月广西医科大学第二附属医院63例RHD患者的右心耳组织,RHD并发心房颤动患者30例作为心房颤动组,RHD合并窦性心律患者33例作为窦性心律组。分别采用RT-PCR、FQ-PCR和Western blot检测HCN2及HCN4的基因及蛋白表达水平,采用Spearman分析两者的相关性。结果RTPCR显示心房颤动组HCN2和HCN4 mRNA表达量均明显高于窦性心律组(t=8.587、13.213,P<0.01)。FQ-PCR显示心房颤动组HCN2和HCN4 mRNA表达量均明显高于窦性心律组(t=10.016、9.105,P<0.01)。Western blot显示心房颤动组HCN2和HCN4蛋白表达量均明显高于窦性心律组(t=13.658、8.590,P<0.01)。心房颤动组HCN2与HCN4 mRNA和蛋白水平上调率均呈现正相关作用(r_(RT-PCR)=0.636,P_(RT-PCR)<0.01;rFQ-PCR=0.683,P_(FQ-PCR)<0.01;r_(wb)=0.641,P_(wb)<0.01)。结论心房颤动组HCN2与HCN4在基因及蛋白表达水平明显高于窦性心律组,且两者的表达水平呈现协同表达,在RHD合并心房颤动的发生发展过程中可能起着重要的作用。 Ojective To explore the coordinated expression of hyperpolarization-activated cyclic nucleotide-gated channel(HCN)2 and HCN4 in atrial fibrillation patients with rheumatic heart disease(RHD).Methods The right atrial appendage tissues of 63 patients with RHD in the Second Affiliated Hospital of Guangxi Medical University from January 2019 to December 2020 were collected.Thirty patients with RHD complicated with atrial fibrillation were included in the atrial fibrillation group,and 33 patients with RHD complicated with sinus rhythm were included in the sinus rhythm group.The gene and protein expression levels of HCN2 and HCN4 were detected by RT-PCR,FQPCR and Western blot,respectively,and the correlation between HCN2 and HCN4 was analyzed by Spearman.Results RT-PCR results showed that the levels of expressions of HCN2 and HCN4mRNA in atrial fibrillation group were higher than those in the sinus rhythm group(t=8.587,13.213;P<0.01).FQ-PCR results showed that the levels of expressions of HCN2 and HCN4 mRNA in atrial fibrillation group were higher than those in the sinus rhythm group(t=10.016,9.105;P<0.01).Western blot results showed that the protein expressions of HCN2 and HCN4 in atrial fibrillation group were higher than those in the sinus rhythm group(t=13.658,8.590;P<0.01).In atrial fibrillation group,the up-regulation rates of HCN2 and HCN4 m RNA and protein levels were both positively correlated(r_(RT-PCR)=0.636,P_(RT-PCR)<0.01;rFQ-PCR=0.683,P_(FQ-PCR)<0.01;r_(wb)=0.641,P_(wb)<0.01).Conclusion The gene and protein expression levels of HCN2 and HCN4 in atrial fibrillation group were significantly higher than those in sinus rhythm group,and their expression levels showed coordinated expression,which may play an important role in the occurrence and development of RHD complicated with atrial fibrillation.
