一种枯草芽孢杆菌的计数方法

摘要 为了对一株高度黏连的产多种酶的枯草芽孢杆菌进行有效的活菌计数,试验采用普通培养基,蛋白胨添加质量分数1%、0.5%,牛肉粉和牛肉膏添加质量分数0.3%,氯化钠添加质量分数1%、0.5%,琼脂粉添加量1.5%、3.0%,在不同添加量下进行活菌计数及芽孢计数研究分析。试验结果表明:普通培养基平板蛋白胨、牛肉粉和牛肉膏、氯化钠不同添加量条件下,该菌株枯草芽孢杆菌在平板显示菌落个数仍然比较大,菌落黏连在一起,造成计数偏低,难以形成有效的活菌计数,对产品质量无法准确判断。针对这种情况,最终优化出一种合适平板培养基,蛋白胨1%、牛肉膏0.3%、氯化钠0.5%、琼脂粉3.0%(均为质量分数),使本菌能够生长并菌落独立,计数结果准确,重复性好。说明在活菌计数和芽孢计数中,优化培养基是某些特殊菌株(菌落黏连)的理想计数方法。 In order to count a highly adherent bacillus subtilis which can produce multiple enzymes,in the experiment,the viable bacteria count and spore count were studied by adding 1%,0.5%peptone,0.3%beef powder and beef extract,1%or 0.5%sodium chloride,1.5%or 3.0%agar powder.The results showed that the number of colonies of Bacillus subtilis was still relatively large on the plate with the common medium containing peptone,beef powder,beef extract and sodium chloride,and the colonies adhered together,resulting in a low count,it was difficult to form an effective count of viable bacteria,and the quality of products could not be accurately judged.In view of this situation,a suitable plate culture medium was finally optimized,containing peptone 1%,beef extract 0.3%,sodium chloride 0.5%and agar powder 3.0%.It was shown that the optimum medium was an ideal method for counting some special strains(colony adhesion)in the enumeration of living bacteria and spores.
