摘要
目的:探讨桃红化浊丸对HSC-T6大鼠肝星状细胞增殖和凋亡的影响。方法:制备桃红化浊丸含药血清,体外培养HSC-T6细胞。将细胞分为空白组(CON),阳性药组(PD),桃红化浊丸高剂量组(THZ-H),桃红化浊丸低剂量组(THZ-L),采用细胞增殖检测(CCK-8)法检测各组细胞增殖情况,流式细胞仪检测各组细胞凋亡情况,蛋白免疫印迹试验(Western blot)检测各细胞中α-SMA、CollagenⅠ、TGF-β1、Smad2蛋白的表达;实时荧光定量聚合酶链式反应(Real-time Polymerase Chain Reaction)检测各组细胞中α-SMA、CollagenⅠ、TGF-β1、Smad2 mRNA的表达情况。结果:与空白组相比,桃红化浊丸能抑制HSC-T6细胞增殖能力,诱导细胞凋亡,明显降低HSC-T6细胞中α-SMA、CollagenⅠ、TGF-β1、Smad2的表达量。结论:桃红化浊丸可能通过抑制TGF-β1/Smad2通路,抑制HSC-T6细胞增殖,促进其凋亡,从而发挥抗纤维化作用。
Objective:To explore the effect of Taohong Huazhuo Pill for HSC-T6 cells on proliferation and apoptosis.Methods:The drug-containing serum of Taohong Huazhuo Pill was prepared and HSC-T6 cells were cultured in vitro.A blank group(CON),a positive drug group(PD),A Taohong Huazhuo Pill high-dose group(THZ-H)and a Taohong Huazhuo Pill low-dose group(THZ-L)were set up to intervene HSC-T6 cells.CCK-8 method was used to detect the proliferation of HSC-T6 cells and flow cytometry was to detect the cell apoptosis in each group.The expression ofα-SMA,CollagenⅠ,TGF-β1,and Smad2 protein in each group was detected by Western blot.Real-time PCR was used to detect the expression ofα-SMA,CollagenⅠ,TGF-β1,and Smad2 mRNA in each group.Results:Compared with the blank controlled group,Taohong Huazhuo Pills could suppress the proliferation of HSC-T6 cells,and the apoptosis was significantly increased in the Taohong Huazhuo Pill high-dose group.Meanwhile,the expression ofα-SMA,CollagenⅠ,TGF-β1,Smad2 in HSC-T6 cells was reduced significantly.Conclusion:Taohong Huazhuo Pill may suppress the proliferation of HSC-T6 cells and induce cells apoptosis by inhibiting the TGF-β1/Smad2 pathway,thereby exerting an anti-fibrosis effect.
