摘要
泛素特异性蛋白酶28(USP28)是去泛素化酶USP家族成员,在真核细胞的细胞周期、细胞凋亡和DNA损伤修复等生命活动中扮演了重要的生理角色。USP28在多种肿瘤中过度表达,是近年来去泛素化蛋白酶家族中新发现的抗肿瘤药物靶标。一些USP28的选择性抑制剂已经显现出潜在的肿瘤靶向治疗前景。本文综述了近年来USP28的结构、功能、抑制剂开发及与重大疾病发生发展关系等方面的研究进展。
Ubiquitin specific protease 28(USP28),a member of the USP family,plays a pivotal physiological role in cell cycle,apoptosis,DNA damage repair and other life activities in eukaryotic cells.USP28 is overexpressed in a variety of tumors and is a newly discovered potential drug target for cancer therapy in recent years.Some selective inhibitors of USP28 have shown potential for targeted cancer therapy.In this manuscript,we reviewed the structure,function,inhibitor development of USP28 and its relationship with the major malignant disease.
