长链非编码RNA NBR2在乳腺癌患者血清中的表达水平及临床意义 被引量:1

Expression and clinical significance of serum long non-coding RNA NBR2 in breast cancer patients

摘要 目的分析血清长链非编码RNA(LncRNA)NBR2的表达在乳腺癌组与健康体检组之间的差异是否具有统计学意义,检测LncRNA NBR2的表达水平对评估罹患乳腺癌风险的价值及其与临床相关指标的分析。方法用实时荧光定量PCR法检测2022年5-12月蚌埠医学院第一附属医院收治的31例未予手术和放化疗治疗的乳腺癌患者血清,另选取同期蚌埠医学院第二附属医院31名健康体检者的血清作为对照,分析乳腺癌组与对照组LncRNA NBR2表达的差异,并分析乳腺癌患者血清中LncRNA NBR2的表达水平与年龄、肿块大小、组织学分级、TNM分期、淋巴结转移、雌激素受体(ER)、孕激素受体(PR)、人表皮生长因子受体2(HER-2)及Ki67的差异,采用ROC曲线分析LncRNA NBR2的表达水平对诊断乳腺癌是否具有临床价值。结果乳腺癌组血清LncRNA NBR2的相对表达水平[0.985(0.488,1.004)]明显低于对照组[1.224(1.087,1.847)],差异有统计学意义(U=174,P<0.001)。LncRNA NBR2的表达水平与患者年龄、ER、HER-2、Ki67无关(P>0.05),而与肿块大小、组织学分级、TNM分期、淋巴结远处转移及PR有关(P<0.05)。ROC曲线分析显示,LncRNA NBR2的ROC曲线下面积为0.771(95%CI:0.651~0.891),当最佳cut-off值为0.548时,LncRNA NBR2诊断乳腺癌的灵敏度为74.2%,特异度为80.6%。结论LncRNA NBR2可能是乳腺癌的一个潜在生物标志物,检测其在血清中的表达有助于乳腺癌的早期筛查及患病风险评估。 Objective To analyze whether the difference of serum LncRNA NBR2 expression was statistically significant between the breast cancer group and the healthy body test group,and detect the value of LncRNA NBR2 expression level in evaluating the risk of breast cancer and analyze its correlation with clinical indicators.Methods The serum of 31 breast cancer patients without operation,radiotherapy and chemotherapy admitted to the First Affiliated Hospital of Bengbu Medical College from May 2022 to December 2022 were detected by real time PCR;the serum of 31 normal people in the same period was taken as the control group to analyze the difference of LncRNA NBR2 expression between breast cancer patients and the control group.To found the difference between the expression level of LncRNA NBR2 in serum of breast cancer patients and various clinical indicators.The receiver operating characteristic(ROC)curve was used to analyze whether the expression level of LncRNA NBR2 had clinical value in the diagnosis of breast cancer.Results GraphPad Prism 8.0.2 software was used to analyze the relative expression level of LncRNA NBR2 in serum of breast cancer group[0.985(0.488,1.004)]was significantly lower than that of healthy control group[1.224(1.087,1.847)],and the difference was statistically significant(U=174,P<0.001).No significant differences were found among the relative expression level of serum LncRNA NBR2 in patients with patient's age,ER,HER2,Ki67(P>0.05),but the differences of relative expression level of LncRNA NBR2 in patients with tumor size,histological grade,TNM stage,distant lymph node metastasis and PR were related(P<0.05).The ROC curve analysis showed that the area under ROC curve of LncRNA NBR2 was 0.771(95%CI:0.651-0.891),and the sensitivity and specificity of LncRNA NBR2 were 74.2%and 80.6%,respectively,when the optimal cut-off value was 0.548.Conclusion LncRNA NBR2 might be a potential biomarker for breast cancer,and detection of its expression level in serum might be helpful for early screening and risk assessment of breast cancer.
