芹菜素对丙烯腈致大鼠睾丸损伤的干预作用 被引量:1

Intervention effects of apigenin on testicular injury in rats exposed to acrylonitrile

摘要 目的探讨芹菜素(apigenin,AP)对丙烯腈(acrylonitrile,ACN)引起的大鼠睾丸损伤具有的干预作用和可能的机制,为ACN的生殖毒性防护和AP的利用提供一定依据。方法60只SPF级成年雄性SD大鼠,随机分为对照组(玉米油)、ACN组(46 mg/kg·bw ACN)、低AP组(46 mg/kg·bw ACN+117 mg/kg·bw AP)、中AP组(46 mg/kg·bw ACN+234 mg/kg·bw AP)、高AP组(46 mg/kg·bw ACN+351 mg/kg·bw AP),每组12只,进行灌胃染毒,1次/d,6 d/周,连续4周,末次称重后处死大鼠,摘取双侧睾丸和附睾,称重。测定睾丸组织中酸性磷酸酶(acid phosphatase,ACP)、碱性磷酸酶(alkaline phosphatase,AKP)、乳酸脱氢酶(lactate dehydrogenase,LDH)和琥珀酸脱氢酶(Succinate dehydrogenase,SDH)活力,丙二醛(malondialdehyde,MDA)、谷胱甘肽(glutathione,GSH)含量,超氧化物歧化酶(superoxide dismutase,SOD)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)、过氧化氢酶(Catalase,CAT)活力及总抗氧化能力(total antioxidation capability,T-AOC)水平。结果与对照组比较,ACN组大鼠附睾湿重和其脏器系数降低,差异有统计学意义(P<0.05);大鼠睾丸组织中AKP、SDH活力降低,差异有统计学意义(P<0.05);GSH-Px活力和T-AOC水平降低,差异有统计学意义(P<0.05)。与ACN组比较,中和高剂量AP干预组大鼠染毒后体重、体重增加量、体重增长率降低,差异有统计学意义(P<0.05);低AP组大鼠附睾湿重和其脏器系数升高,差异有统计学意义(P<0.05);低、中和高剂量AP组大鼠睾丸组织中LDH活力降低,AKP活力升高,差异有统计学意义(P<0.05);低AP组大鼠睾丸组织中GSH含量和T-AOC水平增高,差异有统计学意义(P<0.05);高AP组GSH-Px活力降低,差异有统计学意义(P<0.05)。结论ACN可诱导大鼠睾丸组织发生损伤,其机制可能与脂质过氧化有关;117~234 mg/kg·bw AP对ACN诱导的大鼠睾丸损伤有一定改善作用。 Objective To explore whether the testicular injury induced by acrylonitrile(ACN)would be intervented by apigenin(AP)and its possible mechanisms to provide a certain basis for the protection of reproductive toxicity of ACN and the use of AP.Methods Sixty SPF-grade male rats were randomly divided into the control group(corn oil),ACN group(46 mg/kg·bw ACN),low-dose AP group(46 mg/kg·bw ACN+117 mg/kg·bw AP),middle-dose AP group(46 mg/kg·bw ACN+234 mg/k·bwg AP)and high-dose AP group(46 mg/kg·bw ACN+351 mg/kg·bw AP),12 rats each group.These rats were administered by gavage,once a day,six days a week.After four weeks,all rats were weighed before they were excuted to pick their bilateral testis and epididymis,which were also weighed.Then the activity of acid phosphatase(ACP),alkaline phosphatase(AKP),lactic dehydrogenase(LDH),succinic dehydrogenase(SDH),the content of glutathione(GSH)and malondialdehyde(MDA)were measured in testicular tissue,as well as the activity of superoxide dismutase(SOD),glutathione peroxidase(GSH-Px),catalase(CAT)and the total antioxidant capacity(T-AOC).Results Compared with the control group,the wet weight and the organ coefficient of epididymis in ACN group were decreased significantly(P<0.05).The activity of AKP and SDH in testicle tissue of rats were decreased significantly(P<0.05).And the activity of GSH-Px and T-AOC in testicular tissue of rats were decreased significantly(P<0.05).Compared with the ACN group,the body weight after poisoning,the body weight gain and the body weight growth rate of the rats in the middle and high dose AP groups were significantly decreased(P<0.05).And the wet weight and the organ coefficient of epididymis in low-dose AP rats were significantly increased(P<0.05).The LDH activity in the testicular tissue of the low,middle and high dose AP rats was significantly reduced,and the AKP activity was significantly increased(P<0.05).In the low-dose AP group,the levels of GSH and T-AOC in testicular tissue were significantly increased(P<0.05),and the activity of GSH-Px in high-dose AP group was significantly decreased(P<0.05).Conclusion The testicular injury in rats would be induced by ACN,and its mechanism may be related to lipid peroxidation.The testicular injury in rats induced by ACN would be improved by 117 mg/kg·bw^234 mg/kg·bw AP.
