基于数据挖掘和网络药理学探析中医药治疗冠心病作用机制 被引量:2

Analysis of the Mechanism of Chinese Medicine in Treating Coronary Heart Disease Based on Data Mining and Network Pharmacology

摘要 目的通过数据挖掘类文献,总结中医药治疗冠心病的高频药物,并利用网络药理学分析高频中药可能的关键靶点及作用机制。方法检索知网、万方、维普数据库中药复方治疗冠心病的数据挖掘类文献,分析数据挖掘对冠心病的研究情况,并通过累计频数分析得出前10位高频用药。利用网络药理学分析高频用药治疗冠心病的潜在机制。结果本研究共纳入56篇文献,分析后得出高频用药:丹参、川芎、黄芪、甘草、瓜蒌、当归、茯苓、半夏、薤白、红花。网络药理学分析得出137个共同靶点,主要涉及积极调控细胞增殖、氧化还原过程等生物过程,膜筏、线粒体、突触等细胞结构,酶结合、类固醇激素受体活性、血清素结合等分子功能以及神经活性配体-受体相互作用、钙信号通路、HIF-1等信号通路。包括IL2、NOS2、VCAM1、MPO、TNF、PTGS2、APP等31个关键靶点。结论中医治疗冠心病重在补气、活血、祛痰等方法,活血多用丹参、川芎、红花,补气多用黄芪,祛痰多以瓜蒌、薤白、半夏为主,高频中药不同搭配作用不同,其作用可能与IL2、NOS2、VCAM1、MPO等靶点相关,通过多组分、多通路发挥作用。 Objective Through data mining literature,summarize the high-frequency drugs of TCM for the treatment of coronary heart disease,and use network pharmacology to analyze the possible key targets and mechanism of high-frequency TCM.Methods The data mining literature of Chinese herbal compound treatment of coronary heart disease in the databases of CNKI,WanFang and VIP was searched,and the research status of data mining on coronary heart disease was analyzed,and the top 10 high-frequency medications were obtained through cumulative frequency analysis.Use network pharmacology to analyze the potential mechanism of high-frequency medication for coronary heart disease.Results A total of 56 documents were included in this study.After analysis,it was concluded that high-frequency medications were:Danshen(Salviae Miltiorrhizae),Chuanxiong(Chuanxiong Rhizoma),Huangqi(Astragali Radix),Gancao(Glycyrrhizae Radix Et Rhizoma),Gualou(Trichosanthis Fructus),Danggui(Angelicae Sinensis Radix),Fuling(Poria),Banxia(Pinelliae Rhizoma),Xiebai(Allii Macrostemonis Bulbus),Honghua(Carthami Flos).Network pharmacology analysis revealed 137 common targets,mainly related to the active regulation of cell proliferation,redox process and other biological processes,cell structures such as membrane rafts,mitochondria,synapses,enzyme binding,steroid hormone receptor activity,and serotonin binding other molecular functions and neural active ligand-receptor interaction,calcium signaling,HIF-1 and other signaling pathways.Including 31 key targets such as IL2,NOS2,VCAM1,MPO,TNF,PTGS2,and APP.Conclusion TCM treatment of coronary heart disease focuses on the methods of invigorating Qi,promoting blood circulation,and expectorating.For blood circulation,Gualou(Trichosanthis Fructus),Xiebai(Allii Macrostemonis Bulbus)and Banxia(Pinelliae Rhizoma)are the main methods for eliminating phlegm are more commonly used;Huangqi(Astragali Radix)is more commonly used to invigorate Qi;The role is different,and its role may be related to targets such as IL2,NOS2,VCAM1,MPO,etc.,acting through multiple components and multiple pathways.
