蒙医温针对疲劳大鼠血清细胞因子影响 被引量:3

Effects of Warm Acupuncture on Serum Cytokines in Fatigued Rats

摘要 目的细胞因子在慢性疲劳性疾病的治疗中起着关键性作用。本研究采用疲劳模型大鼠,通过观察温针干预前后IL-1β、IL-4、IL-6、IL-10、TNF-α、IFN-γ的变化,初步阐明温针对疲劳大鼠免疫反应的作用机制。方法清洁级SD大鼠随机分为正常组,正常+温针组,模型组,模型+温针组,每组10只。模型组大鼠采用21 d负重(5%体质量)游泳方法制造运动性疲劳模型。温针组使用MLY-Ⅰ型蒙医疗术温针仪进行温针干预,选取2个穴位,每隔1天交替治疗1次。正常组不给任何处理,平行饲养。之后观察大鼠力竭游泳时间、水迷宫实验等行为学变化以及ELISA法测定IL-1β、IL-4、IL-6、IL-10、血清IFN-γ和TNF-α的含量变化。结果与正常对照组对比,模型组大鼠力竭游泳时间明显缩短,水迷宫总路程明显缩短。血清IL-1β、IL-6、TNF-α含量明显增加(P<0.05或P<0.01)。IL-10含量减少,但无统计学意义。和模型组相比,模型+温针组大鼠力竭游泳时间明显增加,水迷宫总路程明显变长。血清IL-1β、IL-6、TNF-α含量明显变低(P<0.05或P<0.01)。相比模型组,正常+温针组血清IL-1β、IL-6、TNF-α含量明显偏低(P<0.05或P<0.01),IL-10含量明显偏高(P<0.05)。结论运动性疲劳会导致体内血清细胞因子分泌异常,温针能对运动性疲劳大鼠体内异常分泌的细胞因子产生影响,调节紊乱的免疫功能,发挥抗疲劳作用。 Objective:Studies have shown that cytokines play a key role in chronic fatigue syndrome(CFS).In this study,rats with fatigue model were used to observe the changes of IL-1β,IL-4,IL-6,IL-10,TNF-α and IFN-γ before and after Mogolian warm acupuncture intervention.The mechanism of Mongolian warmacupuncture against immune response in fatigue rats was preliminarily elucidated.Methods:SD rats in 4 groups[control group,normal Mongolian warm acupuncture(NMWA)group,model group,model Mongolian warm acupuncture(MMWA)group],10 rats in each group.Rats in the model group were subjected to a 21-day weight(5% body weight)swimming method to create a sports fatigue model.The warm acupuncture group was treated with MLY-I electrothermal acupuncture apparatus for Mongolian warm acupuncture intervention.Two acupoints were selected and treated alternately every other day.The control group did not give any treatment and was raised in parallel.Then,the behavioral changes such as exhaustion time,water maze test and ELISA were used to measure the changes of IL-1β,IL-4,IL-6,IL-10,serum IFN-γ and TNF-α.Results:Compared with the control group,the swimming time and the total distance of the water maze in the model group were significantly shortened.Serum levels of IL-1β,IL-6 and TNF-α were significantly increased(P<0.05 or P<0.01).IL-10 levels were reduced(P>0.05).Compared with the model group,the exhaustive time of the MMWA group increased significantly,and the total distance of the water maze was significantly longer.The levels of serum IL-1β,IL-6 and TNF-α were significantly lower(P<0.05 or P<0.01).Compared with the model group,the levels of serum IL-1β,IL-6 and TNF-α in the MMWA group were significantly lower(P<0.05 or P<0.01),and the IL-10 was significantly higher(P<0.05).Conclusion:Exercise-induced fatigue can cause abnormal secretion of serumcytokines in the body,and Mogolian warm acupuncture can affect abnormal secretion of cytokines in in exercise-induced fatigue rats,regulate disordered immune function,and exert anti-fatigue effects.
