摘要
目的观察脑出血(ICH)灶周组织中白介素-4(interleukin-4,IL-4)、白介素-10(interleukin-10,IL-10)这两种炎症因子的表达及分析其相关意义。从病理学角度分析IL-4、IL-10是否参与(ICH)灶周组织的炎症反应。方法选取5例来源于哈尔滨医科大学第一临床医院神经病学教研室脑库(ICH)死亡患者全脑标本,自出血灶边缘1~3cm及对侧相应部位脑组织取材,对侧设为对照组。通过苏木精-伊红染色观察(ICH)后脑组织的形态学变化,通过免疫组织化学染色观察IL-4和IL-10的表达情况。结果经苏木精-伊红染色及免疫组织化学染色后,(ICH)灶周组织病理改变较明显,有散在的小软化灶,神经元细胞高度水肿,星形胶质细胞聚集,血管外有炎性细胞浸润。出血灶对侧(健侧)神经元核显示不清,部分星形胶质细胞无明显改变。免疫组织化学染色显示(ICH)后脑组织中胶质细胞和神经元细胞胞质内出现棕黄色颗粒状物质,IL-4阳性表达水平较病灶对侧(健侧)高(P<0.05)。观察到距离出血灶越远,IL-4阳性表达越少。而IL-10仅一例病例标本阳性表达,余标本无阳性表达。表达水平与距离出血灶的距离无明显相关。结论IL-4可在(ICH)早期脑组织中表达。(ICH)灶周组织中IL-10的表达可能具有延迟的特点。
Objective To observe the expression of interleukin-4(IL-4)and interleukin-10(IL-10)in the tissues of intracranial hemorrhage(ICH)and analyze their significance.Pathological analysis was performed to determine whether IL-4 and IL-10 were involved in the inflammatory response of the tissues around the focal ICH.Methods Five whole brain specimens from patients with ICH from the brain bank of the Department of Neurology,the First Clinical Hospital of Harbin Medical University were selected.The brain tissues were collected from the edge of the hemorrhage focus 1-3 cm and the corresponding part of the contralateral side,and the contralateral side was set as the control group.The morphological changes of braintissue after intra ICH were observed by hematoxylin-eosin(HE)staining,and the expression of IL-4 and IL-10 was observed by immunohistochemical staining.Results After HE staining and immunohistochemical staining,the pathological changes of the ICH were obvious.There were scattered small softened lesions,neuron cells were highly edema,astrocytes aggregated,and inflammatory cells infiltrated outside the blood vessels.The neuron nucleus on the contralateral side(the healthy side)of the hemorrhage was not clear,and some astrocytes did not change significantly.Immunohistochemical staining showed that brown-granular particles appeared in the cytoplasm of glial cells and neuronal cells in the brain tissue after intra(ICH),and the positive expression level of IL-4 was higher than that on the contralateral side(the healthy side)of the lesion(P<0.05).It was observed that the further away from the bleeding site,the less the expression of IL-4 was.However,only one case of IL-10 was positively expressed,and the other specimens had no positive expression.There was no significant correlation between the expression level and the distance from the bleeding site.Conclusions IL-4 can be expressed in early brain tissue of(ICH).The expression of IL-10 in the perihematomal brain tissue may be delayed.
