摘要
目的探讨麦门冬汤抗肺癌作用及对肿瘤相关巨噬细胞(TAMs)表型的影响。方法建立Lewis小鼠肺癌移植瘤模型,不同浓度麦门冬汤灌胃干预3周,绘制肿瘤增殖曲线并计算抑瘤率;ELISA法检测移植瘤组织中白细胞介素(IL-10、IL-12)的表达。体外用IL-4诱导RAW264.7细胞为M2表型后麦门冬汤干预,流式细胞术检测M2、M1特征性膜分子CD206和CD16/32的表达,观察麦门冬汤对TAMs表型的影响;用IL-4或IL-4+麦门冬汤干预的RAW264.7细胞条件培养基共培养A549细胞,transwell和克隆形成实验检测不同处理条件下A549细胞的迁移和增殖能力,评价麦门冬汤通过调节TAMs表型对肺癌A549细胞的迁移和增殖能力的影响。结果麦门冬汤体内有明确的抑瘤作用,抑瘤率达33.58%,与模型组相比差异具有统计学意义。移植瘤组织中麦门冬汤组较模型组IL-10表达减低,IL-12增高,差异具有统计学意义;体外实验中,流式细胞术检测IL-4组CD206+细胞数明显升高,IL-4+MMDT组CD16/32+表达升高,差异有统计学意义;transwell和克隆形成实验中,RAW264.7+IL-4+MMDT组细胞迁移数量、克隆形成数量均较RAW264.7+IL-4组减少,差异差异有统计学意义。结论麦门冬汤可通过调节TAMs表型抑制细胞增殖、转移。
Objective To explore the anti-lung cancer effect of Maimendong Decoction(麦门冬汤)and its influence on the phenotype of tumor-associated macrophages(TAMs).Methods LLC-transplanted mouse models were established.Different concentrations of Maimendong Decoction intragastric intervention for 3 weeks.The tumor proliferation curve was drawn and the tumor inhibition rate was calculated.ELISA method was used to detect the expressions of interleukin(IL-10 and IL-12)in transplanted tumor tissues.In vitro experiment,after induction of M2 phenotype in RAW264.7 cells with IL-4,Maimendong Decoction was used.Flow cytometry detected the expression of M2,M1 characteristic membrane molecules CD206 and CD16/32 to observe the effect of Maimendong Decoction on the phenotype of TAMs.A549 cells were co-cultured with IL-4 or IL-4+Maimendong Decoction in the conditioned medium of RAW264.7 cells.Transwell and clone formation experiments were used to detect the migration and proliferation ability of A549 cells under different treatment conditions,and evaluate the ability of Maimendong Decoction to regulate proliferation and metastasis of A549 cells based on the regulation of TAMs phenotype.Results Maimendong Decoction had a clear anti-tumor effect in vivo,with a tumor-inhibition rate of 33.58%,which was statistically significant compared with the model group.Compared with those of the model group,the expression of IL-10 in the Maimendong Decoction group in the transplanted tumor tissue was lower,and the expression of IL-12 was higher.The difference was statistically significant.In vitro,flow cytometry detected that the number of CD206+cells in the IL-4 group was significantly increased,and the expression of CD16/32+in the IL-4+Maimendong Decoction group was increased,and the difference was significant.In transwell and clone formation experiments,RAW264.7+IL-4+number of cell migration and the number of clone formation in the Maimendong Decoction group were lower than those in the RAW264.7+IL-4 group,and the difference was statistically significant.Conclusion Maimendong Decoction can inhibit cell proliferation and metastasis by regulating the phenotype of TAMs.
