响应面法优化超高压提取天麻中酚类成分工艺 被引量:4

Optimization of Ultra-High Pressure-assisted Extraction of Polyphenols from Gastrodiae Rhizoma by Response Surface Methodology

摘要 目的:优化天麻中天麻素和对羟基苯甲醇的超高压辅助提取工艺。方法:以乙醇体积分数、料液比、提取压力和保压时间4个影响因素为自变量,天麻中天麻素和对羟基苯甲醇的提取总量为响应值,在单因素试验基础上,采用Box-Behnken中心组合试验,以4因素3水平的响应面分析法,优化天麻超高压辅助提取的工艺条件。结果:最佳提取工艺条件为提取溶剂82%乙醇、料液比1:40、提取压力500 MPa和保压时间4 min,在此条件下,天麻中天麻素和对羟基苯甲醇的实际提取总量为718.59μg·g^-1,预测值为731.27μg·g^-1,相对误差为1.73%。结论:超高压提取法提取天麻中天麻素和对羟基苯甲醇具有高效、简便、耗能少的优点,可用于天麻及其制剂的质量控制过程中活性成分的提取。 Objective:To optimize the ultra-high pressure-assisted extraction of gastrodin and p-hydroxybenzyl alcohol from Gastrodiae Rhizoma.Methods:Ethanol content,solid-liquid ratio,extraction pressure and time of maintain pressure were selected as the independent variables,meanwhile,the total yields of gastrodin and p-hydroxybenzyl alcohol were studied as the response value.Based on single-factor test,the optimum extraction technology was established through Box-Behnken center composite design and the method of response surface analysis with 4 factors and 3 levers.Results:The optimum extraction conditions were obtained as follows:ethanol content of 82%,solid-liquid radio of 1:40,extraction pressure of 500 MPa,and extraction time of 4 min.Under this conditions,the total yields of gastrodin and p-hydroxybenzyl alcohol were 718.59μg·g-1,the relative error was 1.73%compared with the predicted value of 731.27μg·g-1.Conclusion:The extraction of gastrodin and p-hydroxybenzyl alcohol by ultra-high pressure-assisted extraction from Gastrodia elata has advantages of high efficiency,simplicity and low consumption,which can be used for the extraction of active components in the quality control of Gastrodiae Rhizoma and its preparations.
