摘要
体外培养的原代心肌细胞具有良好的自发节律性和收缩性,且不受神经、体液等因素的影响,被认为是研究和治疗心血管系统疾病的稳定而有效的研究对象。传统方法培养的心肌细胞活力、纯度不高,且获得的细胞数量少,因此国内外诸多学者对心肌细胞的培养方法进行研究和改进,旨在建立便捷、高效、稳定的培养方法,以获得存活率、纯度及活性较高、结构功能完整、生长状态好的心肌细胞。目前原代心肌细胞的培养方法纷繁各异,笔者拟就心肌细胞培养方法的研究进行综述,且着重对心肌细胞分离、纯化、培养等方面进行介绍,以期为初次接触原代心肌细胞培养的研究者提供技术支持。
Primary cardiomyocytes cultured in vitro has good spontaneous rhythm and contractility,and is not affected by nerves,body fluids and other factors.It is considered to be a stable and effective research object for the study and treatment of cardiovascular diseases.The cardiomyocytes cultured by traditional methods have low vitality and purity,and the number of cells obtained is small.Therefore,many scholars at home and abroad have researched and improved the culture methods of cardiomyocytes to establish a convenient,efficient and stable culture method to obtain cardiomyocytes with higher viability,purity and activity,complete structure and function,and good growth status.At present,there are many different cultural methods of primary cardiomyocytes.The author intends to review the research on primary cardiomyocytes culture methods,and focuses on introducing separation,purification,and culture of cardiomyocytes to provide technical support for researchers who first contact with culture of primary cardiomyocytes.
