摘要
目的:建立同时测定大鼠体内的甘草次酸和丹参酮含量的RP-HPLC方法,为甘草次酸和丹参酮复方制剂的体内药动学研究提供方法学基础。方法:采用Waters Symmetry C_(18)(250mm×4.6mm,5μm)色谱柱,以甲醇-磷酸缓冲液(pH=3)(85∶15)为流动相进行等度洗脱;体积流速:1.0mL/min;进样量:10μL;柱温:30℃;检测波长:250nm(甘草次酸)、270nm(丹参酮)。结果:甘草次酸和丹参酮的线性范围分别为0.3125μg/mL~20μg/mL(线性回归方程为Y=0.4307X-0.0026,r2=0.9998);0.078125μg/mL~5μg/mL(线性回归方程为Y=2.2377X-0.0222,r^(2)=0.9996);甘草次酸和丹参酮在线性范围内线性良好。低、中、高三种浓度的提取回收率在95.20-103.43%之间,RSD值均小于4.93%,提取回收率良好。该方法在保留时间内未见其他干扰峰,其专属性及精密度均良好。结论:该方法可用于同时测定体内甘草次酸和丹参酮的含量。
Objective:To establishing an RP-HPLC method for the simultaneous measurement of glycyrrhetinic acid and tanshinone in rats,and to provide a theoretical framework for the in vivo pharrmacokinetic study of glycyrrhetinic acid and tanshinone in multiple component formulations.Methods:The isocratic elution was performed on a Waters Symmetry C18 column(250 mm X 4.6 mm,5μm)with methanol-phosphate buffer solution(pH=3)(85:15)as mobile phase;volume flow rate:1.0 mL/min;injection volume:10μL;column temperature:30℃;detection wavelength:250 nm(glycyrrhetinic acid),270 nm(tanshinone).Results:The linear ranges of glycyrrhetinic acid and tanshinone were respectively 0.3125μg/mL~20 pug/mL(the linear regression equation was Y=0.4307X-0.0026,r^(2)=0.9998),0.078125μg/mL~5μg/mL(the linear regression equation was Y=2.2377X-0.0222,r^(2)=0.9996);The mass concentration of glyeyrrhetinic acid and tanshinone showed good linearity with the peak area in the linear range.The extraction recoveries of the three concentrations-low,medium,and high-were between 95.20%and 103.43%,and the RSD values were less than 4.93%,which implies that the extraction recovery rate of the two components is good.There are no other interference Peaks at the corresponding retention time of the main peak in the chromatogram obtained by this method,which indicates that its specificity is good.Conclusion:The method can be used for simultaneous determination of glycyrrhetini cacid and tanshinone in biological samples in vivo.
