摘要 目的:探讨杜仲叶总黄酮对去卵巢骨质疏松大鼠肠道菌群的影响。方法:将32只雌性SD大鼠随机分为假手术组、模型组、雌二醇组(阳性对照)和杜仲叶总黄酮组。采用双侧卵巢摘除法建立绝经后骨质疏松症大鼠模型。用双能X射线骨密度仪测定骨密度;无菌提取粪便DNA,利用Illumina Miseq平台进行高通量测序。结果:与假手术组相比,模型组大鼠体重显著增加(P<0.01),骨密度显著降低(P<0.001),肠道菌群多样性未见明显变化,厚壁菌门丰度降低,拟杆菌门丰度增加;与模型组相比,杜仲叶总黄酮组大鼠体质量增加有所降低,但无显著性差异,骨密度显著增加(P<0.001),肠道菌群多样性未见明显变化,厚壁菌门丰度增加,拟杆菌门丰度降低。结论:实验周期内杜仲叶总黄酮能提高大鼠骨密度,并抑制体重增加,其作用机制可能与调节肠道菌群结构有关。 Objective:To investigate the effects of total flavonoids Eucommia ulmoides leaves on intestinal flora in ovariectomized osteoporotic rats.Methods:32 female SD rats were randomly divided into sham operation group,model group,estradiol group(positive control)and total flavonoid Eucommia ulmoides leaves group.The rat model of postmenopausal osteoporosis was established by bilateral ovariectomy.The bone mineral density of the left femur was measured by dual-energy X-ray absorptiometry.Fecal DNA was aseptically extracted and high-throughput sequencing was carried out by Illumina Miseq platform.Results:Compared with the sham operation group,the body weight of rats in the model group increased significantly(P<0.01),the bone mineral density decreased significantly(P<0.001),the diversity of intestinal flora did not change significantly,the abundance of thick-walled bacteria decreased and the abundance of Bacteroides increased.Compared with the model group,the body weight gain of rats in the total flavonoids group decreased,but there was no significant difference,and the bone mineral density increased significantly(P<0.001),the diversity of intestinal flora did not change significantly,the abundance of thick-walled bacteria increased and the abundance of Bacteroides decreased.Conclusion:Experimental cycle total flavonoids Eucommia ulmoides leaves can improve the rat bone density,and curb weight gain,its mechanism may be related to adjust the structure of intestinal flora.
