摘要
目的探讨人脐带间充质干细胞(hUC-MSCs)对人胰腺癌细胞侵袭能力的调控作用及机制。方法使用含10%胎牛血清的DMEM/F12培养基处理胰腺癌细胞MIA-PaCa-2,并将其设为对照组;使用含10%胎牛血清的人脐带间充质干细胞培养基(hUC-MSC-CM)处理胰腺癌细胞MIA-PaCa-2,并将其设为研究组。Transwell实验评估细胞侵袭力,Western blot实验检测核因子κB(NF-κB)及磷酸化NF-κB(p-NF-κB)水平,分析hUC-MSCs调控胰腺癌细胞侵袭能力的机制。结果Transwell结果显示,对照组MIA-PaCa-2细胞侵袭数为(401±12)个,低于研究组的(715±36)个,差异有统计学意义(P<0.05)。Western blot结果显示,与对照组比较,研究组的NF-κB无明显变化(P>0.05);与对照组比较,研究组的p-NF-κB明显下调(P<0.05)。结论hUC-MSCs可能通过激活NF-κB提高了胰腺癌细胞的侵袭能力。
Objective To investigate the role and mechanism of human umbilical cord-mesenchymal stem cells(hUC-MSCs)in invasive of human pancreatic cancer cells.Methods Pancreatic cancer cells MIA-PaCa-2 were treated with DMEM/F12 medium containing 10%fetal bovine serum and set as the control group.Pancreatic cancer cells MIA-PaCa-2 were treated with human umbilical cord-mesenchymal stem cell medium(hUC-MSC-CM)containing 10%fetal bovine serum and set as the experimental group.The invasive ability were detected by Transwell assay.The levels of NF-κB and phosphorylated NF-κB(p-NF-κB)were detected by Western blot,and the mechanism by which hUC-MSCs regulate the invasive ability of pancreatic cancer cells was analyzed.Results The results of Transwell showed that the invasion number of MIA-PaCa-2 cells in the control group was(401±12),which was lower than that in the experimental group(715±36),and the difference was statistically significant(P<0.05).Western blot showed that compared with the control group,the NF-κB of the experimental group had no significant change(P>0.05).Compared with the control group,p-NF-κB in the experimental group was significantly down-regulated(P<0.05).Conclusion hUC-MSCs may improve the invasive ability of pancreatic cancer cells by activating NF-κB.
