摘要
目的新型冠状病毒疫情在全球传播,了解其变异进化规律,为疫情防控提供参考。方法选取公共数据库中的新型冠状病毒全基因组序列,采用生物信息学方法,对病毒基因组进行系统发育和氨基酸突变位点分析。结果系统发育分析显示,子簇1和2包含多个国家和地区的毒株,子簇3以我国东南沿海地区毒株为主,子簇4为欧洲地区毒株。发现新型冠状病毒存在25个稳定突变位点,其中位于orf8∶84、orf3a∶251、orf1ab∶3607、orf1ab∶4714和S∶613与全基因组进化分成的4个分支具有密切联系。其余突变位点与进化树进一步分簇相关,具有形成稳定克隆的潜力,可能成为新的流行株。结论稳定变异有助于新型冠状病毒传播过程中的适应性进化,种内变异的发现有助于了解病毒的演化方向和流行趋势,为新型冠状病毒监测和溯源提供参考依据。但这些稳定变异位点对病毒的传播和致病能力影响尚未得知,亟需更多分子生物学方面的深入研究。
Objective To identify the patterns of evolution of SARS-CoV-2 that is spreading globally in order to contribute to epidemic prevention and control.Methods The annotated genome sequences of SARS-CoV-2 were downloaded frompublicdatabases,the phylogeny was analyzed and amino acid mutations were identified.Results Phylogenetic analysis showed that clades 1 and 2 contained strains from multiple countries and regions,clade 3 was dominated by domestic strains in the Southeast coastal region,and clade 4 contained strains from Europe.Totally 25 stable mutation sites were found in the genome that might be related to the adaptive evolution of the virus.Among them,the sites located at orf8∶84,orf3a∶251,orf1ab∶3607,orf1ab∶4714,and S∶613 were closely related to the four branches that the whole genome had evolved into.The other mutation sites were related to the further clustering of the evolutionary tree,which had the potential to form stable clones and might become new popular strains.Conclusion Stable mutations represent the adaptive evolution of SARS-CoV-2 during dissemination.The discovery of intra-species variation helps to understand the evolution and epidemic trend of the virus and to monitor and trace the virus.However,the impact of such stable mutations on transmission and pathogenic ability needs to be further studies in molecular biology.
