鼠疫耶尔森菌人工气溶胶的沉降与衰亡空气生物学特性研究 被引量:1

摘要 目的研究鼠疫耶尔森菌201株气溶胶的空气生物学特性,为鼠疫防治工作提供理论依据。方法在气溶胶微环境密闭舱内人工发生鼠疫耶尔森菌201株气溶胶,某些组别发生液内填加荧光素钠指示剂,通过空气动力学粒径谱仪、安德森六级采样器、Biosampler采样器对舱内不同时间点气溶胶进行实时监测或采样,随后对采样皿进行鼠疫耶尔森菌活菌培养计数,对采样液进行鼠疫耶尔森菌基因拷贝数qPCR检测以及荧光素钠浓度分光光度法检测,在此基础上综合分析鼠疫耶尔森菌人工气溶胶空气动力学粒径谱及其沉降规律与衰亡规律。结果发生的鼠疫耶尔森菌气溶胶总粒子空气动力学质量中值直径(MMAD)为2.35μm,生物粒子数量中值直径(NMD)为2.8μm,90%总粒子沉降时间T90为607 min,90%生物粒子衰亡时间T90为2.5 min。结论鼠疫耶尔森菌201株作为气溶胶可被人体吸入至下呼吸道或肺泡,可长期稳定存在于密闭空间内数小时,但会在几分钟内失去活性。 Objective To investigate the aerobiological characteristics of aerosolized Y.pestis strain 201,and provide reference for the prevention and treatment of plague.Methods Y.pestis strain 201 was mixed with or without the sodium fluorescein indicator before being nebulized in an airtight aerosol testing cabin.The Y.pestis aerosol in the cabin was monitored or sampled at different time points by the aerodynamic particle size spectrometer-Anderson six-stage sampler and Biosampler respectively.The CFU of Y.pestis that was collected on the sampling plate was determined by plate counting,the gene copies of Y.pestis in the sampling solution were quantified by qPCR,and the concentration of sodium fluorescein in the sampling solution was measured by spectrophotometry.Then,such aerobiological characteristics of Y.pestis aerosol as the aerodynamic particle size spectrum,deposition and decay characteristics were analyzed synthetically based on the results mentioned above.Results The mass median aerodynamic diameter(MMAD)of the total particle of Y.pestis aerosol was2.35μm,the count median aerodynamic diameter(NMD)of the biological particle of Y.pestis aerosol was 2.8μm,the deposition time of 90%total particles was 607 min,and the decay time of 90%biological particles was 2.5 min.Conclusion Y.pestis aerosol can be inhaled deep into the human respiratory tract and alveoli.F.pestis aerosol can also exist in an airtight space for several hours,but will lose its bioactivity within a few minutes.
