摘要
目的探究β-乳香酸对口腔鳞状细胞癌细胞增殖、迁移和侵袭的作用及可能机制。方法将口腔鳞状细胞癌CAL-27细胞分为Control组、β-乳香酸组、β-乳香酸+空载体组和β-乳香酸+USP22组,CCK-8检测细胞增殖,Transwell检测细胞迁移和侵袭,qRT-PCR检测细胞CREB1和USP22 mRNA表达,Western blot检测细胞CREB1和USP22蛋白表达。结果β-乳香酸抑制CAL-27细胞增殖、迁移和侵袭,抑制CREB1和USP22mRNA和蛋白表达,过表达USP22逆转β-乳香酸对CAL-27细胞增殖、迁移和侵袭的抑制作用。结论β-乳香酸通过抑制CREB1/USP22信号通路抑制口腔鳞状细胞癌细胞增殖、迁移和侵袭。
Objective To explore the effect and possible mechanism ofβ-boswellic acid on proliferation,migration and invasion of oral squamous cell carcinoma cells.Methods The CAL-27 cells were divided into control group,β-boswellic acid group,β-boswellic acid+empty vector group andβ-boswellic acid+USP22 group.Cell proliferation was detected by CCK-8 assay,and cell migration and invasion were detected by transwell assay.The expression of CREB1 and USP22 mRNA were detected by qRT-PCR,and the expression of CREB1 and USP22 proteins were detected by Western blot.Resultsβ-boswellic acid inhibited the proliferation,migration and invasion of CAL-27 cells,inhibited the expression of CREB1 and USP22 mRNA and proteins,and overexpression of USP22 reversed the inhibitory effect ofβ-boswellic acid on the proliferation,migration and invasion of CAL-27 cells.Conclusionβ-boswellic acid inhibits proliferation,migration and invasion of oral squamous cell carcinoma cells by inhibiting the CREB1/USP22 signaling pathway.
