摘要
目的探讨参芪止疱颗粒对HSV-2感染豚鼠相关炎症因子IL-2、IL-6、IFN-γ、TNF-α、NF-κB表达的影响。方法Dunkin Hartly白化雌性豚鼠随机分为正常组、模型组、参芪止疱颗粒组和阿昔洛韦组,建立HSV-2感染豚鼠生殖道模型,并给予相应药物灌胃治疗。RT-PCR法测定各组豚鼠子宫、卵巢IL-2、IL-6、IFN-γ、TNF-α、NF-κB mRNA的表达情况。结果参芪止疱颗粒组与模型组相比,IL-2mRNA和IFN-γmRNA表达上调(P<0.05),而IL-6m RNA、TNF-αmRNA和NF-KB mRNA表达下调(P<0.05)。结论参芪止疱颗粒可通过调控IL-2、IL-6、IFN-γ、TNF-α、NF-κB mRNA的表达,减轻HSV-2感染豚鼠子宫、卵巢组织的炎性损害,从而保护机体,达到抗病毒的作用。
Objective To explore the effect of Shenqi zhipao granule on the expression of relative inflammatory factors in guinea pig with HSV-2,involving interleukin-2(IL-2),IL-6,interferon-γ(IFN-γ),tumor necrosis factor-α(TNF-α)and nuclear factorκB(NF-κB).Methods Female albino Dunkin Hartly guinea pigs were randomly divided into control group,model group,Shenqi Zhipao granule group and acyclovir group,the genital tract model of guinea pig with HSV-2 was established and corresponding medication was given by gastrogavage for 10 d.mRNA expression of IL-2,IL-6,IFN-γ,TNF-αand NF-κB in uterus and ovary of guinea pigs were tested by RT-PCR.Results Compared with model group,the expression level of IL-2 mRNA and IFN-y mRNA in Shenqi Zhipao granule group increased(P<0.05),and the expression level of IL-6 mRNA,TNF-αmRNA and NF-κB mRNA decreased(P<0.05).Conclusion Shenqi Zhipao granule can reduce the inflammatory damage of infected uterus and ovary tissues so as to protect the body and achieve the function of anti-virus by regulating relative inflammatory factors,involving IL-2,IL-6,IFN-γ,TNF-αand NF-κB.
