摘要
目的分别应用两种体系对一株乙酸钙不动杆菌(简称Ac)的致病性进行评估。方法按照《微生物肥料生物安全通用技术准则》(NY 1109—2017)致病性实验进行致病性评估,试验方法包括急性经口毒性试验、急性腹腔注射致病性试验、溶血试验、抗菌药物敏感实验、一次破损皮肤刺激试验、急性眼刺激试验;用秀丽隐杆线虫进行致病性评估,方法主要为寿命试验,此外还研究了Ac对线虫的发育、运动、摄食、消化功能、繁殖的影响。结果雌性、雄性小鼠经口LD50均大于10 g/kg,属实际无毒;急性腹腔注射致病性试验结果显示,试验期间动物未出现中毒体征,无死亡,大体解剖未见异常;溶血试验结果为阴性;Ac对7种不同类别的抗菌药物均敏感,仅对一种药物耐药;Ac对家兔破损皮肤和眼均无刺激性。应用线虫体系发现,Ac对线虫寿命基本无影响,此外,Ac对线虫纵向横断面积、头部摆动频率、身体弯曲频率基本无影响,使咽泵频率增加,平均排便循环时间缩短,虽使线虫后代数目减少,但仍能正常繁殖和产卵。结论两种体系均显示,该乙酸钙不动杆菌菌株无致病性。
Objective To evaluate the pathogenicity of a strain Acinetobacter calcoaceticus(Ac)by two systems.Methods Pathogenicity test was done based on General Biosafety Standard for Microbial Fertilizer(NY1109-2017),including acute oral toxicity test,pathogenic test of acute intraperitoneal injection,hemolytic test,drug susceptibility test,one-time damaged skin irritation test and acute eye irritation test;The method for pathogenicity evaluation using Caenorhabditis elegans was mainly lifespan test,and the effects of Ac on development,movement,feeding,digestion,and reproduction of nematodes were also studied.Results The oral LD50 both for male and female mice was more than 10 g/kg,which was practically non-toxic.The pathogenicity test of acute intraperitoneal injection indicated that no signs of poisoning were showed in the animals during the experiment,with no death and no abnormalities in gross anatomy;The hemolysis test result was negative;Ac was sensitive to seven different antibacterial drugs,only resistant to one drug;Ac was non-irritating to damaged skin and eyes of rabbits.In C.elegans system,the lifespan of the nematodes was unaffected by Ac.In addition,the longitudinal cross-sectional area of the nematodes,the head thrash frequency and the body bends frequency were not affected by Ac.The pharyngeal pump rate was increased and the average defecation cycle time was shortened.Although the brood size was reduced,the nematodes could still fertilize and lay eggs.Conclusion Both systems show that this Ac strain is not pathogenic.
