柞蚕核型多角体病毒侵染对不同柞蚕品种幼虫中肠SOD及CAT基因表达的影响

Effects of ApNPV Infection on Expression of SOD and CAT Genes in the Midguts of Larvae from Different Tussah Varieties

摘要 柞蚕核型多角体病毒(ApNPV)是柞蚕血液型脓病的病原物,柞蚕血液型脓病严重影响柞蚕产量,是制约柞蚕产业经济效益增长的主要因素之一。为探究病原物侵染后柞蚕的抗逆机制,分别对4个柞蚕品种(辽蚕527、辽蚕582、抗大、9906)的幼虫添食ApNPV,分析侵染后12 h、24 h、36 h、48 h、60 h、72 h抗氧化酶基因ApSOD和ApCAT mRNA相对表达量的变化趋势。结果显示,4个柞蚕品种的幼虫中肠ApSOD基因相对表达量在感染ApNPV后24 h开始上调,36 h达到最高值,随后下调;ApCAT基因的相对表达量在感染后12 h开始上调,在24~36 h达到峰值,随后迅速下降。2个基因相对表达量总体呈现先上升后下降的变化趋势,且4个品种的处理组在侵染前期相对表达量显著高于对照组,而后期显著低于对照组。研究结果可为探究柞蚕对ApNPV感染的生化响应机制提供理论依据。 Antheraea pernyi nucleopolyhedrovirus(ApNPV)is the pathogen of Antheraea pernyi nucleopolyhedrosis,which seriously affects the yield of tussah and is one of the main factors restricting the growth of economic benefit of the tussah industry.In order to investigate the resistance mechanism of tussah after pathogen infection,ApNPV was fed to the larvae of four varieties(Liaocan 527,Liaocan 582,Kangda and 9906),and the trends of ApSOD and ApCAT gene mRNA relative expression level at 12 h,24 h,36 h,48 h,60 h,and 72 h after infection were measured.The results showed that the relative expression level of ApSOD in the larvae of the four tussah varieties was up-regulated at 24 h after infection with ApNPV,reached the highest level at 36 h,and then was down-regulated;the relative expression level of ApCAT gene was up-regulated at 12 h after infection,reached the peak at 24-36 h,and then rapidly de-creased.The relative expression levels of the two genes showed a general trend of increasing and then decreasing,and the relative expression levels of two genes of the four varieties were significantly higher than those of the control groups at the early stage of infection but significantly lower than those of the control group at the later stage.The results can provide a theoretical basis for investigating the biochemical response mechanism of tussah to ApNPV infection.
