摘要
利用植物组织培养技术可以快速获得大量保持品种优良遗传特性的种苗,故适用于以异花授粉为主的桑树优良品种的规模化繁殖与推广。以优良果桑品种德果1号的腋芽作为外植体,以MS和1/2 MS为基础培养基,添加不同浓度的植物生长调节剂6-BA(6-benzylaminopurine)、2,4-D(2,4-dichlorophenoxyacetic acid)、IBA(indole-3-butyric acid)、NAA(α-naphthylacetic acid)进行该品种的组织培养繁殖试验。结果表明:桑芽外植体最佳灭菌条件为75%乙醇消毒30 s+20 g/L Na Cl O消毒8 min;正交试验优化的桑芽增殖培养基为MS+1.4 mg/L 6-BA+0.03 mg/L 2,4-D+0.08 mg/L IBA,最适生根培养基为1/2 MS+0.8 mg/L NAA。上述试验结果有助于优良果桑品种徳果1号的快速繁殖和大面积推广应用。
Tissue culturing technique can quickly obtain a large number of seedlings with good genetic characteristics and it is suitable for large-scale proliferation and popularization of cross-pollinated mulberry.Taking axillary buds of the excellent fruit mulberry variety Deguo 1 as explants,MS and 1/2 MS as basal medium,different concentrations of plant growth regulators 6-BA(6-benzylaminopurine),2,4-D(2,4-dichlorophenoxyacetic acid),IBA(indole-3-butyric acid)and NAA(α-naphthylacetic acid)were used for tissue culturing and proliferation experiment.The results showed that the optimal sterilization condition of mulberry bud explants was 75%ethanol for 30 s and 20 g/L Na Cl O for 8 min.Orthogonal experiment result showed that the optimal basal medium for mulberry bud proliferation was the combination of MS,1.4 mg/L 6-BA,0.03 mg/L 2,4-D and 0.08 mg/L IBA,and the best rooting medium was 1/2 MS with 0.8 mg/L NAA.The above experimental results are helpful for the rapid proliferation and large-scale application of the excellent fruit mulberry variety Deguo 1.
