摘要
目的:探究建立高脂饮食诱导的高血脂大鼠模型,为降脂药物的筛选和疗效评价提供适宜的实验模型。方法:将18只SPF级SD雄性大鼠(体重180~220g)分为2组,以正常饲料喂食为对照组(10只),以高脂饲料喂食为模型组(8只)。在造模第1周和第4周分别测定大鼠血清中TC、TG、HDL-C、LDL-C的含量,并在造模第8周观察大鼠肝脏组织经HE染色的病理切片。结果:模型组大鼠高脂饲料喂养1周后,4项血脂指标(TC、TG、HDL-C、LDL-C)未发生显著变化;继续喂养3周后,模型组大鼠血清TC、TG、LDL-C含量均显著高于正常对照组,而HDL-C的含量显著低于正常对照组;第8周时模型组大鼠的肝脏病理切片与正常对照组比较,肝细胞质内大量空泡,肝细胞核被挤到边缘,肝细胞呈现弥漫性脂肪变性。结论:用高脂饲料喂养大鼠4周后,大鼠出现非常明显的血脂异常,可建立比较稳定的高血脂模型,此方法可用于高血脂实验动物模型的建立。
Objective:To establish a rat model of hyperlipemia,and provide an animal experimental model for screening lipid-lowering drugs and evaluating the curative effect.Method:Eighteen SPF-grade SD male rats(weight 180-220 g)were divided into 2 groups,fed with normal chow as the control group(10 rats)and fed with high-fat chow as the model group(8 rats).The levels of TC,TG,HDL-C and LDL-C in the serum of rats were measured at the 1 st and 4 th weeks of modeling;and the pathological sections of liver tissues stained with hematoxylin-eosin were observed at the 8 th week of modeling.Results:After the model group rats were fed with high-fat diet for 1 week,the four lipid indexes(TC,TG,HDL-C,LDL-C)did not change significantly;after 3 weeks of feeding,the serum TC,TG and LDL-C levels of the model group rats were significantly higher than those of the normal control group,while the level of HDL-C was significantly lower than that of the normal control group;at week 8,the liver pathological sections of the model group rats were compared with those of the normal control group,and the liver The hepatocytes showed a large number of vacuoles in the cytoplasm,the nuclei of hepatocytes were squeezed to the side,and the hepatocytes showed diffuse steatosis.Conclusions:After feeding rats with high-fat chow for 4 weeks,rats can show very obvious dyslipidemia,and a more stable hyperlipidemia model can be established,and this method can be used for the establishment of experimental animal models of hyperlipidemia.
