三阴性乳腺癌中FOXC1,HIF-1α及上皮间质转化因子的表达及意义 被引量:2

摘要 目的:探讨FOXC1、缺氧诱导因子-1α(HIF-1α)在三阴性乳腺癌(TNBC)中的表达及与上皮间质转化(EMT)免疫表型的相关性及其临床意义。方法:回顾性分析78例TNBC患者临床病理资料及随访结果。对这78例TNBC标本及30例癌旁正常乳腺组织采用免疫组化染色法检测FOXC1,HIF-1α及上皮间质转化相关因子E-cadherin、Vimentin蛋白的表达,分析其与患者临床病理参数的关系,探讨FOXC1,HIF-1α的表达与上皮间质转化的关系。结果:FOXC1,HIF-1α在TNBC中的表达阳性率分别为53.8%、60.3%与癌旁正常乳腺组织中表达的差异具有统计学意义(P<0.05);FOXC1蛋白的表达与肿瘤的淋巴结转移有关(P<0.05),HIF-1α蛋白的表达与组织学分级及淋巴结转移有关(P<0.05)。FOXC1,HIF-1α的表达与E-cadherin的低表达和Vimentin的高表达存在相关性。结论:TNBC组织中FOXC1,HIF-1α的检测对预测肿瘤侵袭转移、判断预后具有一定价值。 Objective:To investigate the expression of FOXC1 and hypoxia-inducible factor-1α(HIF-1α)in triple negative breast cancer(TNBC)and its correlation with epithelial-mesenchymal transition(EMT)immunophenotype and its clinical significance.Methods:The clinical and pathological data of 78 patients with TNBC were retrospectively analyzed,with the expression of FOXC1,HIF-1αand E-cadherin and Vimentin protein detected by immunohistochemical staining in 78 cases of TNBC specimens and 30 cases of adjacent normal breast tissues,and the pathological parameters and the relationship among FOXC1,HIF-1αand EMT analyzed.Results:The positive expression rates of FOXC1 and HIF-1αin TNBC were 53.8%and 60.3%respectively,higher than those in normal breast tissues(P<0.05).The expression of FOXC-1 protein was significantly correlated with lymph node metastasis(P<0.05).The expression of HIF-1αprotein was significantly correlated with differentiation degree and lymph node metastasis(P<0.05).The expression of FOXC1 and HIF-1αwas correlated with the low expression of E-cadherin and the high expression of Vimentin.Conclusion:The detection of FOXC1 and HIF-1αmay be valuable in predicting tumor invasion and metastasis in TNBC tissues.
