摘要
以污染的蛹虫草栽培料为试材,采用划线分离法分离病原真菌,并对其进行形态学观察及ITS序列系统发育分析,以期鉴定人工栽培蛹虫草栽培料中污染真菌的种类。结果表明:从蛹虫草栽培料中分离到3株真菌菌株,P1和P5分离物与GenBank中多株深绿木霉分离物同源性最高,均在99.6%以上,结合形态学特征,鉴定P1和P5分离物为深绿木霉。分离物P3和扩展青霉同源性最高,结合形态特征,鉴定P3为扩展青霉。该研究结果为人工栽培蛹虫草真菌病害的鉴别与防治提供了参考依据。
In order to identify the species of contaminated fungi in cultured Cordyceps militaris medium,the pathogenic fungi was separated by streak plate method,and morphological identification and ITS sequence phylogenetic analysis were carried out.The results showed that three fungi strains were isolated from the contaminated culture material during C.militaris cultivation.P1 and P5 isolates had the highest homology with several Trichoderma atroviride isolates in GenBank,all of which were more than 99.6%.Combined with morphological characteristics,P1 and P5 isolates were identified as T.atroviride.The strain P3 had the highest homology with Penicillium expansum isolates in GenBank,and combined with morphological characteristics,P3 was identified as P.expansum.The results provided a theoretical basis for the identification and control of the fungus diseases of C.militaris cultivation.
