摘要
目的:探讨曲古霉素A(TSA)对脂多糖(LPS)诱导肺泡巨噬细胞炎症反应的影响。方法:体外培养大鼠肺泡巨噬细胞株NR8383,分为4组:磷酸盐缓冲液(PBS)对照组、LPS(1μg/mL)处理组、TSA(1 ng/mL)预处理30 min+LPS(1μg/mL)处理组、TSA(10 ng/mL)预处理30 min+LPS(1μg/mL)处理组。各组细胞处理后继续培养6 h,ELISA法检测细胞培养上清液肿瘤坏死因子α(TNF-α)蛋白水平,实时定量PCR法检测细胞TNF-αmRNA表达。结果:与PBS对照组比较,LPS处理组细胞培养上清液TNF-α蛋白水平明显升高,细胞TNF-αmRNA的表达量升高(8.68±0.56)倍(均P<0.05)。与LPS处理组比较,TSA(1 ng/mL和10 ng/mL)处理组细胞培养上清液TNF-α蛋白水平和细胞TNF-αmRNA的表达量均明显降低(均P<0.05)。结论:TSA预处理可抑制LPS诱导的大鼠肺泡巨噬细胞炎症反应。
Objective:To investigate the effect of trichostatin A(TSA)on lipopolysaccharide(LPS)induced inflammatory response of the alveolar macrophages.Methods:Rat alveolar macrophages(cell line NR8383)were culture in vitro,and then randomly divided into four groups:control group(stimulated with phosphate buffer solution),LPS-stimulated group(stimulated with 1 ng/mL LPS),TSA-treated groups(30 min pretreatment with 1 ng/mL or 10 ng/mL TSA,then stimulated with 1 ng/mL LPS).After incubation for 6 h,the cells were harvested and the supernatant of cell culture medium was collected.The levels of TNF-αprotein in the supematant were assayed by using enzyme-1 inked immunosorbent assay(ELISA),and the expression of TNF-αmRNA of cells was detected by using real-time quantitative PCR.Results:Compared with control group,the supernatant level of TNF-αprotein was significantly higher,and the expression of TNF-αmRNA in NR8383 cells was increased by 8.68±0.56 folds(bothP<0.05).Compared with LPS-stimulated group,the protein and mRNA expressions of TNF-αwere significantly reduced in TSA-treated groups(1 ng/mL and 10 ng/mL)(allP<0.05).Conclusion:TSA pretreatment may inhibit LPS induced inflammatory response in rat alveolar macrophages.
