摘要
蛋白磷酸酶2A(protein phosphatase 2A,PP2A)有着逆转蛋白激酶的作用,对调控磷酸化信号的通路有重要作用。因细胞周期蛋白依赖激酶(cyclin-dependent protein kinases,CDKs)在精子发生的过程有着重要的功能,而PP2A能通过去磷酸化功能影响CDK的活性,即PP2A可能对精子发生的过程产生影响,进而推测PP2A与雄性犏牛不育有关。为了验证PP2A是否对精子的发生产生影响,在本研究中对犏牛睾丸PP2A基因进行克隆得到其CDs区序列,并通过生物信息学法对其分析,同时利用Q-PCR技术检测PP2A基因在牦牛和犏牛的睾丸及附睾中的表达情况。结果表明:犏牛PP2A基因的CDs区编码长度为930 bp,由309个氨基酸组成,是带负电的酸性亲水稳定蛋白;同偶蹄类的同源性较高,在系统进化树中距离较近;不是分泌蛋白而是结合转运蛋白,无跨膜的结构域。牦牛PP2A在睾丸和附睾中的表达都极显著高于犏牛(P<0.01),犏牛的PP2A在睾丸和附睾中的表达下调,推测其可能会对某些重要的信号通路造成一定影响并进一步作用于犏牛精子的发生过程,即PP2A与雄性犏牛不育有关。
Protein phosphatase 2 A(PP2 A)has the function of reversing protein kinase,and plays a certain role in regulating the phosphorylation signal pathway.Because cyclin-dependent protein kinases(CDKs)play an important role in the process of spermatogenesis,PP2 A is capable of influencing the activity of CDK through dephosphorylation,namely,PP2 A may have an effect on the process of spermatogenesis,and thus PP2 A is speculated to be related to male infertile of cattleyak.In order to verify whether PP2 A is involved in the development of spermatogenesis and has an impact on it,the CDs region sequence of PP2 A was cloned from the testes of cattle-yak in this study,and bioinformatics analysis was carried out on it.Meanwhile,Q-PCR was used to detect the expression of PP2 A gene in the testes and epididymis of yak and cattle-yak.The results showed that the CDs region of cattle-yak PP2 A gene had an encoding length of 930 bp,and the 309 amino acid sequences encoded were conservative,with negative acid hydrophilic stable protein.The homology with artiodactyla was higher and the distance was closer in phylogenetic tree.It was not a secretory protein but a binding transporter and had no transmembrane domain.The expression of PP2 A in yak was significantly higher in both testis and epididymis than in cattle-yak(P<0.01),and the expression of PP2 A in the testis and epididymis of cattle-yak was down-regulated,suggesting that the significant down-regulation of PP2 A gene in cattleyak may have certain influences on some important signaling pathways and further contribute to the spermatogenesis of cattle-yak and PP2 A is related to male infertile of cattle-yak.
