聚乙二醇-6000模拟干旱胁迫下硅对蒙古黄芪种子萌发特性和幼苗质量的影响 被引量:5

Effects of silicon on seed germination characteristics and seedling quality of Astragalus membranaceus( Fish) Bge.var.mongholicus( Bunge.) Hsiao under PEG-6000 simulated drought stress

摘要 目的研究干旱胁迫下硅对蒙古黄芪种子萌发特性和幼苗质量的影响,探索硅在提高蒙古黄芪抗旱性方面的作用。方法采用二因素完全随机设计及培养皿沙培试验,以聚乙二醇(PEG)-6000模拟不同程度的干旱胁迫,并加入硅(浓度分别为0,3,6 mmoL/L),测定各处理下种子萌发特性和幼苗质量。结果随PEG-6000模拟浓度的增加,各测定指标均呈不同程度的降低。1)重度胁迫下发芽率、发芽势和发芽指数较正常供水分别降低48.7%,76.1%,75.1%,硅可明显缓解以上指标的下降趋势;2)重度胁迫下全株长、子叶长、子叶宽、鲜重较正常供水和中度胁迫明显降低(P<0.05),中度胁迫与正常供水比较差异无统计学意义(P>0.05),而硅的加入除全株长外,对其他指标均无明显影响;3)中度胁迫下叶片相对含水量较正常供水降低3.5%,重度胁迫较中度胁迫和正常供水分别降低7.6%,10.8%,6 mmoL/L的硅可使重度胁迫下的叶片相对含水量明显升高(P<0.05);4)在重度胁迫下,叶绿素a含量和叶绿素a/b值较正常供水分别降低13.6%,28.0%,叶绿素b变化不明显,硅可使叶绿素含量呈不同程度的增加。结论适当干旱胁迫对蒙古黄芪种子萌发和幼苗生长无显著影响,重度胁迫可显著抑制种子萌发、幼苗生长和叶绿素的合成;硅能干预重度干旱胁迫对黄芪种子萌发和幼苗生长的抑制作用,为蒙古黄芪的规范化生产和硅肥的合理使用提供了试验依据。 Objective To study the effect of silicon on seed germination characteristics and seedling quality of Astragalus membranaceus(Fish)Bge.var.mongholicus(Bunge.)Hsiao under drought stress,and to explore the role of silicon in improving drought resistance of Astragalus membranaceus(Fish)Bge.var.mongholicus(Bunge.)Hsiao.Methods Two-factor random design and Petri dish sand culture experiment were used to simulate drought stress in different degrees with PEG-6000,and silicon(the concentration was 0,3 and 6 mmol/L,respectively.)was added to determine seed germination characteristics and seedling quality under different treatments.Results With the increase of simulated concentration of PEG-6000,the determinative indexes showed a decreasing trend in varying degrees.1)The germination rate,germination potential and germination index under severe stress were 48.7%,76.1%and 75.1%lower than CK,respectively.Silicon significantly alleviated the downward trend.2)The whole plant length,cotyledon length,cotyledon width and fresh weight of plant under severe stress were significantly lower than those under CK and moderate stress(P<0.05),but there was no significant difference between moderate stress and CK(P>0.05),while the addition of silicon had no significant effects on other indexes except the whole plant length.3)Relative water content of leaves under moderate stress was 3.5%lower than CK,while that under severe stress was 7.6%and 10.8%lower than that under moderate stress and CK,respectively.And 6 mmoL/L of silicon made the relative water content of leaves under severe stress increase significantly(P<0.05).4)Under severe stress,content of chlorophyll a and chlorophyll a/b decreased by 13.6%and 28.0%respectively compared with CK,but chlorophyll b did not change significantly.Silicon increased chlorophyll content in different degrees.Conclusion Appropriate drought stress has no significant effect on seed germination and seedling growth of Astragalus membranaceus(Fish)Bge.var.mongholicus(Bunge.)Hsiao.Severe stress significantly inhibits seed germination,seedling growth and chlorophyll synthesis.Silicon can improve the inhibition of severe drought stress on seed germination and seedling growth of Astragalus membranaceus(Fish)Bge.var.mongholicus(Bunge.)Hsiao,which provides experimental basis for standardized production of Astragalus membranaceus(Fish)Bge.var.mongholicus(Bunge.)Hsiao and rational use of silicon fertilizer.
