丹参酮IIA对帕金森大鼠中脑内Caspase-12和CHOP的影响 被引量:9

Effects of Tanshinone IIA on expression of Caspase-12 and CHOP in the substantia nigra in rats with Parkinson′s disease

摘要 目的观察丹参酮IIA对帕金森病(PD)模型大鼠中脑黑质内CHOP蛋白、Caspase-12蛋白、Caspase-3蛋白和酪氨酸羟化酶(TH)蛋白表达的影响,探讨其对帕金森病模型大鼠的作用及相关机制。方法(1)帕金森病(PD)模型大鼠的制备:60只健康、雄性SD大鼠,采用数字表法随机分为模型组(30只)、假手术组(sham组)(15只)和正常对照组(15只)。对照组对照组正常饲养,不作任何处理;假手术组,采用在大鼠颈背部皮下注射不含鱼藤酮的葵花油乳化液;模型组采用在大鼠颈背部皮下注射鱼藤酮。每天注射1次,连续30 d。用药期间观察实验大鼠外观和行为学变化。(2)把制备成功的模型组先分为处理组(丹参酮IIA组,15只)和生理盐水组(NS组,15只)。两组大鼠分别被注射生理盐水和丹参酮IIA,每天注射1次,连续注射14 d。(3)用药期间观察实验大鼠外观和行为学变化。(4)第14 d,采用免疫组织化学染色检测3组大鼠中脑黑质内TH蛋白、CHOP蛋白、Caspase-12蛋白和Caspase-3蛋白的表达情况。结果与正常对照组及假手术组比较,模型组大鼠出现典型的帕金森病样症状,可以进行后续实验;免疫组织化学染色见NS组大鼠中脑黑质内可见CHOP蛋白、Caspase-12蛋白和Caspase-3蛋白的表达增多,TH蛋白的表达则降低,与正常对照组比较,差异有统计学意义(P<0.05)。处理组大鼠中脑黑质内可见CHOP蛋白、Caspase-12蛋白和Caspase-3蛋白的表达降低,TH蛋白的表达则增多,与NS组比较,差异有统计学意义(P<0.05)。结论丹参酮IIA通过减弱内质网应激反应介导的神经元调亡,从而减轻帕金森病模型大鼠中脑黑质内多巴胺能神经元的丢失。 ObjectiveTo explore the mechanism of Tanshinone IIA on rats with Parkinson′s disease by way ofobserving effects of Tanshinone IIA on expression of tyrosine hydroxylase(TH),CHOP,Caspase-12 and Caspase-3 in thesubstantia nigra.Methods(1)Sixty male adult SD rats were randomly divided into PD model group(n=30),sham group(n=15)and control group.The control group was not given any intervention.PD model group was evenly and randomly dividedinto two groups:treatment group(TSA group)(n=15)and saline group(NS group)(n=15).The PD model group wasestablished by subcutaneous injection of rotenone in the back and neck area,while the injection of sunflower oil emulsionwithout rotenone with the same quantity and at the same point as the PD model group was applied in the sham group.(2)Naturesaline 1 ml and tanshinone IIA 20 mg/kg were given through intraperitoneal injection respectively for 14 days in the PD modelgroup.TH,CHOP,Caspase-12 and Caspase-3 were measured with immunohistochemical staining in the substantia nigra.ResultsPD model results:compared with the control group and sham group,there was typical PD ethologic change in PDmodel group,the follow-up experiments can do.Immunohistochemical staining results:compared with the control group,theexpression of TH positive neuron decreased,while the expression of CHOP,Caspase-12 and Caspase-3 increased significantly in the substantia nigra in NS group.Compared with the NS,the expression of TH positive neuron increased,while theexpression of CHOP,Caspase-12 and Caspase-3 decreased significantly in the substantia nigra in the TSA group.ConclusionTanshinone IIA can mitigate the loss of dopaminergic neurons in the substantia nigra in the Parkinson′s diseasemodel rats induced by rotenone,which may be related with reducing apoptosis mediated by the endoplasmic reticulum stress.
