摘要
目的观察鳖甲煎丸对肝硬化模型大鼠肝功能及血清GLU、TG、TGF-β1、CTGF、PDGF的影响,探讨鳖甲煎丸抗肝硬化糖脂代谢紊乱及肝纤维化的作用机制。方法SD大鼠30只适应性饲养1周后,随机分为空白组10只、造模组20只。空白组自由喂养,造模组采用四氯化碳复合法诱导构建肝硬化门脉高压症大鼠模型大鼠。选择16只造模成功的肝硬化模型大鼠随机分为模型组和鳖甲煎丸组,每组8只。模型组自由喂养,鳖甲煎丸组以中成药鳖甲煎丸0.9 g/kg剂量灌胃干预8周,每日1次。取大鼠肝脏组织和下腔静脉血,肝组织切片后采用苏木精-伊红(HE)及Masson染色电镜下观察肝组织纤维化情况,采用试剂盒检测大鼠血清AST、ALT、TP、ALB、TBil、GLU、TG、TGF-β1、CTGF、PDGF水平。结果(1)空白组大鼠肝细胞结构完整、排列整齐,未见明显细胞变性、坏死;模型组大鼠肝细胞排序混乱,出现明显脂肪变性,肝脏汇管区可见大量胶原纤维增生,小叶结构破坏明显,肝组织出现明显的条索样改变;鳖甲煎丸组大鼠肝组织纤维增生明显改善,仅见少量纤维组织增生,肝小叶结构完整,肝小叶结构破坏得到不同程度修复,小叶间隙缩窄,肝细胞排列规整、未见明显脂肪变性。(2)与空白组比较,模型组AST、ALT、TP、ALB、TBil、GLU、TG、PDGF、TGF-β1、CTGF差异有统计学意义(P<0.01);经过8周的干预,鳖甲煎丸组AST、ALT、TBil、GLU、TG、PDGF、TGF-β1、CTGF显著低于模型组(P<0.01),ALB水平显著高于模型组(P<0.01),TP水平与模型组差异无统计学意义(P>0.05)。结论鳖甲煎丸能显著改善肝硬化模型大鼠的肝功能和糖脂代谢紊乱,作用机制可能与调控PDGF、TGF-β1、CTGF等纤维化细胞因子有关。
ObjectiveTo observe the effect of Biejiajian Pill on liver function and serum Glu,TG,TGF-β1,CTGF,PDGF in cirrhotic model rats,and to explore the mechanism of Biejiajian Pill on the disorder of glycolipid metabolism and liver fibrosis in cirrhotic rats.Methodsafter one week of adaptive feeding,30 SD rats were randomly divided into 10 blank rats and 20 model rats.The rats in the blank group were fed freely,and the model rats were induced by carbon tetrachloride.16 cirrhotic rats were randomly divided into model group(8 rats)and Biejiajian Pill group(8 rats).The rats in the model group were fed freely,and the rats in the Biejiajian Pill group were given a dose of 0.9 g/kg by gavage for 8 weeks,once a day.The liver tissue and inferior vena cava blood of rats were taken.The liver tissue was sectioned and the fibrosis was observed by he and Masson staining electron microscopy.The serum ast,ALT,TP,ALB,TBil,Glu,TG,TGF-β1,CTGF and PDGF levels were detected by kit.Results(1)the structure of hepatocytes in the blank group was complete and arranged orderly,without obvious cell degeneration and necrosis;the hepatocytes in the model group were disorganized,with obvious fatty degeneration;a large number of collagen fibers were proliferated in the portal area of the liver;the structure of lobules was damaged obviously,and the liver tissue showed obvious cord like changes;the proliferation of liver tissue fibers in the Biejiajian Pill group was significantly improved,only A small amount of fibrous tissue hyperplasia was observed,the structure of the liver lobule was complete,the damage of the structure of the liver lobule was repaired to varying degrees,the interlobular space was narrowed,the arrangement of the liver cells was regular,and no obvious cell steatosis was found.(2)There were significant differences in AST,ALT,TP,ALB,TBil,Glu,TG,PDGF,TGF-β1,CTGF between the model group and the blank group(P<0.01).After 8 weeks intervention,AST,ALT,TBil,Glu,TG,PDGF,TGF-β1,CTGF in Biejiajian Pill group were significantly lower than those in model group(P<0.01),ALB level was significantly higher than that in model group(P<0.01),TP level was not significantly different from that in model group(P>0.05).Conclusion Biejiajian Pill can significantly improve liver function and glycolipid metabolism disorder in cirrhotic rats,and its mechanism may be related to the regulation of PDGF,TGF-β1,CTGF and other fibrogenic cytokines.
