摘要
基于高效液相色谱(High Performance Liquid Chromatography,HPLC)技术建立了天麻酒中天麻素含量测定的方法.样品经氮气吹至近干,纯水复溶,并经0.22μm针头式微孔滤膜过滤后,直接上机检测.方法以C_(18)AQ色谱柱(250 mm×4.6 mm×5μm)为分离柱,以乙腈和水作为流动相,利用保留时间定性,外标法定量.研究结果显示,天麻素在1~500 mg/L的浓度范围内线性良好,相关系数r为0.9999;检出限为0.74 mg/kg.在40 mg/kg,80 mg/kg和160 mg/kg的添加水平下,基体加标回收率分别在100.5%~104.2%、99.1%~102.9%和100.2%~102.1%范围内,相对标准偏差分别为0.7%、0.9%和0.6%.该方法操作简单,具有较高的准确度和精密度,可用于天麻酒中天麻素含量的准确测定.
A method for the determination of gastrodin in tianma medicinal liquor was established by high-performance liquid chromatography(HPLC).The sample was near dried by nitrogen stream,and then the residue was redissolved by pure water.The solution was submitted to filtrate via a 0.22μm microporous film and injected into the HPLC system.Chromatographic separations of gastrodin were performed on a C_(18)AQ column(250 mm×4.6 mm×5μm).An isocratic elution program was used with the mobile phase,consisting of acetonitrile and water.In this method,the retention time was used for qualitative analysis,and external standard method was used for quantification.Good linear relationships and coefficients of determination(r=0.9999)were achieved over the concentration range of 1 to 500 mg/L.The limit of detection was 0.74 mg/kg.The recoveries at spiking matrix levels of 40 mg/kg,80 mg/kg and 160 mg/kg were 100.5%-104.2%,99.1%-102.9%and 100.2%-102.1%respectively.The relative standard deviations(RSDs)were 0.7%,0.9%and 0.6%respectively.This developed method showed highly accuracy and precision,and could be used to determine the amount of gastrodin in tianma medicinal liquor.
