摘要
目的 探讨芬太尼通过调节环氧合酶2(Cox?2)对肝癌SMCC?7721细胞增殖、凋亡的影响.方法 培养人肝癌SMCC?7721细胞,MTT实验、流式细胞仪分别检测不同浓度芬太尼(0、0.5、5、50 μg/L)及下调Cox?2对细胞增殖、凋亡的影响,qRT?PCR和免疫印迹检测不同浓度芬太尼对细胞中Cox?2 mRNA和蛋白表达的影响,MTT实验和流式细胞仪检测上调Cox?2对芬太尼处理后细胞增殖、凋亡的影响.结果 MTT实验和流式细胞仪检测结果,芬太尼呈时间、浓度依赖性抑制肝癌细胞增殖,浓度依赖性诱导肝癌细胞凋亡;不同浓度芬太尼处理后,肝癌细胞中Cox?2 mRNA和蛋白表达均明显降低(均P<0.05),呈浓度依赖性;沉默Cox?2可明显抑制肝癌细胞增殖,诱导细胞凋亡;上调Cox?2可逆转芬太尼抑制肝癌细胞增殖、诱导细胞凋亡的作用.结论 芬太尼具有抑制肝癌细胞增殖、诱导细胞凋亡的作用,其作用机制可能与芬太尼抑制Cox?2基因表达有关.
Objective To investigate the effects of fentanyl on proliferation and apoptosis of hepatoccarcinoma cell line SMCC?7721 by regulating cyclooxygenase?2(Cox?2).Methods Human liver cancer SMCC?7721 cells were cultured.MTT assay and flow cytometry were used to detect the effects of different fentanyl doses(0,0.5,5,50μg/L)and down?regulation of Cox?2 on cell proliferation and apoptosis.qRT?PCR and Western blotting were used to detect the effects of different fentanyl doses on Cox?2 mRNA and protein expression in SMCC?7721 cells.Then,MTT assay and flow cytometry were used to determine the effect of Cox?2 up?regulation on cell proliferation and apoptosis after fentanyl treatment.Results MTT assay and flow cytometry showed that fentanyl may inhibit the proliferation of hepatocarcinoma cells in a time?and dose?dependent manner,and induce apoptosis of hepatocarcinoma cells in a dose?dependent manner.After treatment with different doses of fentanyl,the Cox?2 mRNA and protein expression in the hepatocarcinoma cells were significantly decreased(P<0.05)in a dose?dependent manner.Silencing Cox?2 can may significantly inhibit the proliferation and induce apoptosis of the hepatocarcinoma cells.Up?regulation of Cox?2 may regress the effect of fentanyl on inhibiting proliferation and inducing apoptosis of hepatocarcinoma cells.Conclusion Fentanyl may inhibit the proliferation and induce apoptosis of hepatocarcinoma cells.The mechanism may be related to the inhibition of Cox?2 gene expression by fentanyl.
