多靶位基因测序快速鉴定脓肿分枝杆菌复合群 被引量:1

摘要 目的 探讨和评价多靶位基因测序联合分析的方法用于脓肿分枝杆菌复合群的快速菌种鉴定.方法 收集经PNB/TCH鉴别培养基表型鉴定和16S rRNA基因测序鉴定为脓肿分枝杆菌复合群的临床分离株,再应用hsp65和rpoB基因测序的联合分析进行种/亚种的鉴定.结果 经表型鉴定为非结核分枝杆菌的52株临床分离菌株,16S rRNA基因测序比对分析与脓肿分枝杆菌的同源性高达100%.最后经hsp65和rpoB测序及16S rRNA基因序列的联合分析鉴定40株菌株为脓肿分枝杆菌脓肿亚种,10株为脓肿分枝杆菌马赛亚种以及2株脓肿分枝杆菌bolletii亚种.结论 16S rRNA、hsp65和rpoB三个靶位基因测序联合分析的方法能够快速将脓肿分枝杆菌复合群分离株鉴定至亚种的水平. Objective To evaluate the multi-target genes sequencing analysis for rapid identification of Mycobacterium abscessus complex. Methods The clinical isolates were collected and identified as NTM by PNB/TCH differential medium phenotype test. Members Were verified by the sequencing of 16S rRNA gene. The identification of species/subspecies was carried out by using three genes PCR sequencing combined analysis. Results In total,52 clinical specimens were identified as NTM by phenotype test. 16S rRNA gene sequencing analysis of all samples shared over 100%similarity with MABC. Finally,species identification with 16S rRNA,hsp65 and rpoB sequencing analysis revealed that 40 strains were M. abscessus subsp. abscessus,10 were M. abscessus subsp. massiliense and 2 was M. abscessus subsp. bolletii. Conclusion The combined analysis of 16S rRNA, hsp65 and rpoB three genes sequencing can quickly identify the MABC isolates to the subspecies level.
