电针百会 大椎穴对高脂喂养AD兔模型脑组织代谢的影响

摘要 目的 探讨针刺对高脂喂养阿尔茨海默病(AD)模型兔脑组织代谢的影响.方法 将18只日本大白兔(JWR)随机分成三组,对照组(NC)、AD组及电针组(EA)各6只,分别给予12周普通饮食喂养、2%高胆固醇饮食及2%高胆固醇饮食喂养并电针针刺兔'百会'、'大椎'穴,利用MRS技术检测脑组织代谢物浓度,随后处死取脑,采用ELISA法检测脑皮质中Aβ1-42蛋白表达,采用单因素方差分析对各组间代谢物、Aβ1-42表达进行统计分析.结果 AD组的NAA/Cr(0.66±0.07)和?Glu/Cr(0.64±0.06)较NC组(0.81±0.08和0.87±0.06)明显减低,AD组海马Aβ1-42浓度(238.62±58.16)pg/mg较NC组(155.57±26.17)pg/mg明显增高,差异有统计学意义.EA组NAA/Cr(0.78±0.07)和Glu/Cr(0.78±0.09)明显高于AD组,海马Aβ1-42(166.38±30.92pg/mg)浓度明显低于AD组.结论 针刺能减缓高脂喂养AD模型兔脑组织代谢AD样改变的进程. Objective To investigate the effects of acupuncture Baihui and Dazhui points on the brain metabolism of AD rabbit model induced by high cholesterol diet.Methods 18 JWR were randomly divided into 3 groups:normal control group(NC,n=6),AD(n=6)and electro acupuncture group(EA,n=6).Each group was given routine diet,high cholesterol diet and high cholesterol diet with electroacupuncture therapy for 12 weeks,respectively.Proton magnetic resonance spectroscopy(1H MRS)of rabbit brains was performed with 3.0 T MR Scanner to quantify neural metabolites.The levels of Aβ1-42 protein in hippocamus were observed by ELISA postmortem.The differences of the levels of neural metabolites and Aβ1-42 between groups were statistically analyzed by ANOVA.Results The ratio of NAA/Cr(0.66±0.07)and Glu/Cr(0.64±0.06)in group AD were significantly lower than the group NC(0.81±0.08 and 0.87±0.06).The levels of Aβ1-42 protein in group AD(238.62±58.16)pg/mg was significantly higher than the group CD(155.57±26.17)pg/mg.The ratio of NAA/Cr(0.78±0.07)and Glu/Cr(0.78±0.09)in group EA were significantly higher than the group AD(0.66±0.07).The levels of Aβ1-42 protein in group EA(166.38±30.92)pg/mg was significantly lower than the group CD(238.62±58.16)pg/mg.Conclusion Acupuncture can relieve the AD-like changes of brain metabolism in the AD rabbit model induced by high cholesterol diet.
